Development of SCAR Markers Linked to L3 gene in Capsicum

Abstract: Random amplified polymorphic DNA (RAPD) markers linked to the L 3 locus were developed by applying the bulked segregant analysis method to two doubled haploid (DH) populations in Capsicum. The codominant RAPD markers, E18 272 and E18 286 , were converted into sequence-characterized amplified region (SCAR) markers by molecular cloning and nucleotide sequencing. A PCR analysis using DH (n = 176) and backcross (n = 190) populations revealed that all the SCAR markers, PMFR11 269 , PMFR11 283 and PMFR21 200 , co-se… Show more

“…'K9-11' harbors the L 3 gene, which is resistant to pepper mild mottle virus (PMMoV) derived from PI 159236. This non-pungent bell-shaped sweet pepper is susceptible to P. capsici (Yanokuchi et al 1993, Sugita et al 2004. 'AC2258' is highly resistant to P. capsici and is pungent (Smith et al 1967, Yamakawa et al 1979, Palloix et al 1990).…”

“…In the Capsicum species, several linkage maps have been reported (Prince et al 1993, Lefebvre et al 1995, Livingstone et al 1999, Kang et al 2001. Thus, DNA markers linked to various agronomic characters were developed (Caranta et al 1999, Moury et al 2000, Blum et al 2002, Matsunaga et al 2003, Sugita et al 2004. These DNA markers are now available for MAS for some traits.…”

“…'K9-11' harbors the L 3 gene derived from PI159236, and is a bell-like sweet pepper (Yanokuchi et al 1993, Sugita et al 2004. It is susceptible to phytophthora blight and has no pungency, and its immature fruit color is dark green.…”

“…An RFLP marker TG36 using a tomato probe, was mapped at 4.0 cM from L 3 with an interspecific DH population by Lefebvre et al (1995). The SCAR markers, PMFR11 269 , PMFR11 283 and PMFR21 200 were mapped at 4.0 cM from L 3 with an intraspecific DH population by Sugita et al (2004) using a bulked segregant analysis method. A SCAR marker, WA31-1500S, was mapped at 1.5 cM from L 4 with an intraspecific F 2 population by Matsunaga et al (2003).…”
