Development of Reference Procedures for Broth Microdilution Antifungal Susceptibility Testing of Yeasts with Standardized Endpoint Determination

Abstract: Standard guidelines for the broth microdilution antifungal susceptibility testing of amphotericin B, flucytosine, fluconazole, miconazole and itraconazole are reported. These are a modification of the method developed by the National Committee for Clinical Laboratory Standards (NCCLS) on the following two points: standardization of the means of endpoint determination and the inclusion of miconazole and itraconazole in the testing. MIC was determined to be when the positive control had a turbidity of 0.2 at the… Show more

“…Broth micro-dilution assay was performed according to the standard guidelines for the broth micro-dilution susceptibility test for yeast (18) or for filamentous fungus (22), with necessary modifications. The use of 96 flatshaped plastic microplates was avoided because of possible adsorption of water-insoluble components of essential oils on the plastic surface.…”

“…This kit was reported as equivalent to the JSMM standard method (2) and MIC values have been used after 24-hr incubation for almost all of the ascomycotic yeasts, including Candida species except C. glabrata and C. krusei (1,12). NCCLS recommended that 24-hr incubation is also meaningful because 24-hr MIC reading is often possible 104 K. MAKIMURA ET AL and 24-hr readings may be more clinically relevant for some isolates.…”

“…In addition, there has been controversy regarding optimization of the incubation time for antifungal susceptibility testing (8). The JSMM microdilution method and its commercial version, frozen Eiken plates, were developed for reliable comparable antifungal susceptibility testing to the NCCLS M27-P macrodilution method (4) for five antifungal agents in 1994 (1,12). These aims were achieved using our endpoint reading system in which corrections were made for the differences in growth rates of tested cultures.…”
