Development of Rapid Assessment Method to Determine Bacterial Viability Based on Ultraviolet and Visible (UV-Vis) Spectroscopy Analysis Including Application to Bioaerosols

Abstract: We evaluated a method for the assessment of bacterial viability that is based on ultraviolet and visible (UV-Vis) spectroscopy analysis. The quantities of intracellular materials inside a cell vary depending on change of bacterial viability by disruption of the membrane integrity. Therefore, normalized optical density in the range of 200-290 nm was analyzed to determine if it varied in samples containing different proportions of live bacteria. Our results indicate that samples containing higher proportions of … Show more

“…Bioaerosols in the atmosphere pose potential health hazards such as allergy, asthma, bronchitis, dermatitis and diarrhea (Lavoie et al, 2006;Taha et al, 2006;Park et al, 2012). Deleterious health effects are possible even when the bioaerosols are non-viable (Griffiths and DeCosemo, 1994).…”

“…These would have the flexibility for use in combination with various observation or detection techniques: for example, counting and characterization of stained microbes using epifluorescence microscopy (Nakajima et al ; Hara and Zhang ; Murata and Zhang ), and flow cytometry (Chen and Li ; Besmer et al ; Buysschaert et al ). Combinations of fluorescent staining with other methods also have been developed (Bisson et al ; Park et al ; Choi et al ; Yamaguchi et al ; Sorensen et al ; Swanson and Huffman ). Although the use of fluorescent staining has been reported as above, the staining conditions appear to be determined by trial and error.…”

“…These methods include impaction, filtration, impingement and depositional sampling. A rapid assessment method was developed to determine bacterial viability by applying ultraviolet and visible spectroscopy analysis (Park et al, 2012). However, an evaluation of sensitivity, repeatability and accuracy for bioaerosol sampling is important and it is essential to determine the capabilities and limitations of the various available sampling methods.…”
