Development of Powdery Mildew Resistance in Cucumber Using CRISPR/Cas9-Mediated Mutagenesis of CsaMLO8

Abstract: Powdery mildew (PM) diseases may severely limit the production of various crops, including members of to the Cucurbitaceae family. Successful PM infection relies on the Mildew Resistance Locus O (MLO) plant gene family, which encodes susceptibility (S) factors essential for fungus penetration into the host cell. In cucumber, natural mutations in CsaMLO8 confer resistance to the PM pathogen Podosphaera xanthii. Here, we used CRISPR/Cas9-mediated mutagenesis to generate PM resistance in the susceptible cucumber … Show more

“…The symptoms are characterized by the presence of powdery-looking patches or a diffusive layer of white, yellow, brown, or grayish mycelia and conidiophores on the plant surfaces of the plants. This disease is a serious threat to the production of various crops, including those belonging to the Cucurbitaceae family [30,31]. Currently, some biofungicides have been registered and recommended for the control of cucumber powdery mildew disease in many countries [9,32].…”

“…Powdery mildew is one of the devastating diseases of Cucurbitaceae family. Two transgene-free Csamlo8 CRISPR mutant lines (Csamlo-cr-1 and Csamlo-cr-2) showing high resistance to PM were isolated from susceptible cucumber cultivar 'Ilan' using CRISPR/Cas9-mediated mutagenesis (Shnaider et al 2022). Genome editing especially by CRISPR-Cas9 is a cost-effective, highly efficient method due to its targeted approach.…”

“…Powdery Mildew affects the correct development of the plant, severely affecting the quality of the fruit and causing yield losses. Nonetheless, several studies have reported genetic resistance against this disease in several species of cucurbits (He et al, 2013;Wang et al, 2016;Shnaider et al, 2022;López-Martín et al, 2022).…”
