Development of Novel Glucose and Pyruvate Biosensors at Poly(Neutral Red) Modified Carbon Film Electrodes. Application to Natural Samples

Abstract: Amperometric biosensors based on the corresponding oxidase enzyme with poly(neutral red) redox mediator have been developed for the determination of glucose and pyruvate. The enzymes have been immobilized on top of poly(neutral red) modified carbon film electrodes with glutaraldehyde as the cross-linking agent. The biosensors were characterized by cyclic voltammetry and by electrochemical impedance spectroscopy. The glucose biosensor exhibited a linear response in the range 90 mM to 1.8 mM with a detection lim… Show more

“…Poly(neutral red) (PNR) has been extensively used as mediator for the construction of various biosensor assemblies, for glucose, pyruvate and alcohol detection [13,14]. A biosensor was developed for use in flow analysis for glucose monitoring, containing the enzyme entrapped in a sol-gel layer on a PNR-modified carbon film electrode [15].…”

“…Neutral red (NR), an acid base indicator with pK a ~6.8 from the family of azines, can be electropolymerised and poly(neutral red) (PNR) has already been used as redox mediator in enzyme electrodes [17,22]. The combination of different polymers to obtain Page 4 of 29 A c c e p t e d M a n u s c r i p t 4 composites with better properties than each polymer by itself is of current interest [13].…”

“…Glutaraldehyde has usually been used as a protein cross-linking agent in biosensor fabrication using carbon-film electrodes [8][9][10][11][12][13]. Biosensors for glucose determination have been reported using ferrocene [9], CoHCF [10], methyl viologen [11] and PNR [12] as redox mediators, and multienzyme sensors have been used for analysis of various foodstuffs [13]. It is a bifunctional cross-linking agent which reacts with lysine residues on the exterior of the proteins.…”
