Development of Near-Isogenic Line of japonica Rice Cultivar Saenuri without Lipoxygenase-3

Abstract: It is reported that the absence of lipoxygenase-3 (LOX-3) may contribute to a reduction in stale flavor after the storage of rice.To improve the quality of stored rice of the Korean japonica rice cultivar, we conducted a breeding program to develop near-isogenic rice without LOX-3 in the genetic background of Saenuri, a mega variety of Korea. In the first step of the breeding program, we used a donor parent of LOX-3 null, Daw Dam, and a recurrent japonica parent, Sindongjin, to develop HR27873-AC12 by backcros… Show more

“…Among 386 KASP markers showing polymorphism, markers showing hetero genotypes were excluded, and 96 markers located at 5 Mb intervals per 12 chromosomes in rice were selected to analyze the recurrent parent genome recovery ratios in BC 1 F 1 and BC 2 F 1 of Samgwang/Seolgaeng. A genetic graphic map was derived using a MapChart program (version 2.32) based on the physical locations of the KASP markers on chromosomes [ 31 ].…”

“…Ninety-six markers located at a 5 Mb intervals on 12 rice chromosomes were selected to analyze the recovery ratio of the recurrent parent genome in BC 1 F 1 and BC 2 F 1 . Genetic graphical mapping was performed using a MapChart program (version 2.32) based on the physical location of the KASP markers on chromosomes [ 61 ]. The agronomic traits such as plant height, culm length, panicle length, and the number of tillers were phenotyped based on a standard method of RDA, Korea [ 56 ] in the lines selected in the BC 1 F 1 and BC 2 F 1 generations.…”

“…Unkwang was crossed to RSV-resistant variety Haedamssal as the donor of the RSV resistance gene, and the genetic backgrounds of BC 2 F 1 and BC 2 F 2 plants were analyzed with KASP markers to enable selection of a NIL with 96.2% recovery of the recurrent parent genome [ 22 ]. In a separate program to breed rice with reduced stale flavors after storage, 406 KASP markers were used for genetic background analysis of Jeonju624, a NIL of Korean temperate japonica rice variety Saenuri with a null allele of lipoxygenase-3 introduced from donor line HR27873-AC12, and revealed 95.8% recovery of the recurrent parent genome [ 23 ].…”
