Mucoadhesive buccal films of rivastigmine were prepared by the solvent casting technique using HPMC K15M, sodium alginate, glycerine, and Eudragit RL100. Arranged films assessed for weight variation, thickness, % drug substance, % moisture loss, % moisture take-up, folding endurance, in-vitro medicament release, and Fourier transform Infrared spectroscopy (FTIR). The films showed a controlled release (CR) over 8 h. The preparation observed to be a worthy candidate for the development of buccal patches for therapeutic purposes. Drug-polymer compatibility considers FTIR demonstrated no contradiction between the medicament and the polymers. The optimized formulation found F7 indicated drug release 85% at the end of 8 h. Thinking about the correlation coefficient (R2) values got from the kinetic equations, the drug release from the formulations F1-F8 has discovered zero-order release mechanism. It can be concluded that oral buccal patches of rivastigmine, for treatment of Alzheimer’s and Parkinson’s disease, can be formulated. The study suggests that rivastigmine can be conveniently administered orally in the form of buccal patches, with the lesser occurrence of its side effects and improved bioavailability.

Conventional dosage forms for topical and transdermal drug delivery have several disadvantages related mainly to its poor skin permeation and patient compliance. Many approaches have been developed to improve these dosage forms. Film forming drug delivery systems represents a recent advancement in this field. It provides improved patient compliance with enhanced skin permeation of drugs. In its simplest form, these consist of a polymeric solution, usually in a supersaturated state, in a suitable solvent. A plasticizer is usually added to improve the flexibility and enhance the tensile strength to the film. It is also possible to control and sustain the drug release from the films by controlling the polymeric content, concentration of plasticizer, or formulation with other additives. In this review, we are summarizing the mechanism of formation of these films as well as its types and possible applications. The main ingredients, properties, and evaluation of the various film forming delivery systems are also discussed.

Delivery of drugs into the systemic circulation via transdermal drug delivery system is well known. Transdermal drug delivery system is one of the most reliable and convenient delivery system for the delivery across skin. The aim of this research project is to develop a matrix diffusion control transdermal therapeutic system that combines the drug Ramosetron hydrochloride with an amount of hydrophilic and hydrophobic polymers using a solvent extraction method. Ramosetron hydrochloride is a highly potent drug and has an extended duration of action ad minor side effects despite the severity that it undergoes hepatic pass metabolism and therefore the purpose of this study was to prepare a transdermal patch for Ramosetron hydrochloride for the treatment of chemotherapy induced nausea and vomiting (Anti-emetic action) and evaluate it for various parameters.
