Development of IVM‐IVF produced 8‐cell bovine embryos in simple, serum‐free media after conditioning or Co‐culture with buffalo rat liver cells

Abstract: Development of 8-cell bovine embryos derived from in vitro matured/in vitro fertilized (IVM/IVF) oocytes was evaluated in two simple, serum-free media (CZB and SOM) with buffalo rat liver cells co-culture (BRLC) or after conditioning compared to a commonly used, serum-supplemented complex medium TCM-199. In a 3 x 4 factorial design, 578 eight-cell embryos were randomly assigned to 12 treatment groups. The factors were: first, type of culture medium (M199/FBS, CZBg and SOM), and second, the use of BRLC (as co-c… Show more

“…Menezo's B2 medium was used in this experiment because it is one of the most successful chemically defined media for culturing bovine embryos and contains no LIF or serum (Menezo et al, 1984). Although TCM-199 is an ideal medium for BRL cells (Rehman et al, 1994;Reed et al, 1996), it is not a good medium for early-stage bovine embryos in the absence of BRL cells (Hernandez-Ledezma et al, 1993). It was determined that LIF added to Menezo's B2 medium had no effect on percentage of embryos developing into blastocysts in B2 medium with or without serum.…”

“…However, primary cell cultures are often inconvenient and quite variable for use in embryo culture. Buffalo Rat Liver ( BRL) cells, a permanent cell line, have been used successfully for coculture of bovine embryos (Rehman et al, 1994;Van Inzen et al, 1995;Reed et al, 1996).…”

“…These may include conditioning of the medium by secretion of diffusible compounds such as differentiation inhibitory activity (DIA; Smith and Hooper, 1987;Gearing et al, 1987;Smith et al, 19881, removal, or modification of secreted factors or substances already present in the medium. Interaction of the embryonic cells with molecules expressed on the surface of the feeders may also play an important role, though there is no evidence to suggest a species specificity in this interaction, since both mouse ES cells and bovine embryo growth (Rehman et al, 1994) can be supported on BRL cells.…”
