Development of Ic-ELISA and Colloidal Gold Lateral Flow Immunoassay for the Determination of Cypermethrin in Agricultural Samples

Abstract: Cypermethrin (CYP) is an insecticide in the pyrethroid family and is used widely in agriculture and for public health purposes. However, CYP has been shown to have negative impacts on reproduction, immunity and nerves in mammals. In this study, a monoclonal antibody (mAb) against CYP was prepared and used to establish an indirect competitive immunosorbent assay (ic-ELISA) and colloidal gold lateral flow immunoassay (LFIA) for the quantitative and qualitative determination of CYP residues in agricultural produc… Show more

“…The LFA was tested in three different dilutions of blank sera (clotted blood and heat-inactivated) in running buffer (diluted 10×, 20×, and 100×), as it is understood that matrix effects can be reduced by diluting the matrix in buffer 47 . In both blank serum matrices, the 10× dilution resulted in the appearance of both test lines.…”

“…18 LODs in the range from 0.35 mg L −1 to 416.3 mg L −1 were achieved by these methods. [12][13][14][15][16][17][18] Moreover, other approaches for cypermethrin detection gained low LODs, for example, UV-vis spectrophotometry based on hydrolyzed cypermethrin reacting with iron(III) chloride to form ferricyanide complex ions (LOD of 0.023 mg L −1 ), 19 electrochemical sensor based on anatase TiO 2 -carbon paste obtaining LOD of ∼0.1 mg L −1 , 20 uorescence nanosensor related to MIP-CdSe/CdS/ZnS quantum dots (LOD of 0.3 mg L −1 ) 21 and silver nanorods for SERS (LOD ∼0.42 mg L −1 ). 22 Despite the fact that these strategies are highly responsive and specic, they are associated with cumbersome functionalization procedures and sophisticated Raman equipment.…”

“…Cypermethrin has been determined by surface-enhanced Raman scattering (SERS) with AuNPs-zirconia nanofibers, 12 Au/Ag/Au nanorod arrays, 13 AuNPs core–shell molecularly imprinted polymer (MIP), 14 and bacterial nanocellulose decorated with AuNPs. 15 A lateral flow immunoassay strip using AuNPs labeled with mAb 2 G7H9, 16 and an immunochromatographic strip-based sensor using polymer carbon dots have been fabricated. 17 Recently, gold-functionalized magnetic nanoparticle combined with a broad-spectrum antibody CL-CN/1D2 was developed as a sensor for pyrethoids.…”

“…Immunoassays have been widely used to detect various analytes in biomedicine, food, and the environment because of the advantages of being simple, fast, and economical [16]. Among immunoassays, the enzyme-linked immunosorbent assay (ELISA) is widely considered the most prevalent and mature method [17]. To date, indirect competitive ELISAs (ic-ELISAs) on the basis of anti-CAP monoclonal antibody (mAb) for CAP detection in environmental and agricultural samples have shown half-inhibitory concentrations (IC 50 ) of 1.6 ng/mL [18] and 1.5 ng/mL [19].…”

“…However, the long reaction time, tedious operating procedures, and the need for specialized equipment have limited the application of ELISAs in field testing [20]. Compared with ELISAs, gold nanoparticle-based lateral flow immunoassays (AuNP-LFIAs) possess the advantages of one-step detection and do not require specialized equipment and direct visual readout [17]. Therefore, they have tremendous potential for on-site rapid testing and have been used to detect various small-molecule pesticides, such as quizalofop-p-ethyl [20], quinclorac [15], and deoxynivalenol [21].…”
