Development of High-Grade Sarcoma After Second Dose of Moderna Vaccine

Abstract: Coronavirus disease 2019 (COVID-19) is a disease characterized by predominantly respiratory symptoms, which can progress to respiratory failure. Due to the novelty of the vaccines, it is difficult to assess if there are any associated long-term side effects. Here, we present a case of an elderly female who received the Moderna COVID-19 vaccine and developed a high-grade sarcoma at the site of the injection. A 73-year-old female with a past medical history of hypertension, hyperlipidemia, and renal angiomyolipo… Show more

“…Interestingly, there have been reports of the Moderna vaccine being associated with malignancy. One case described a sarcoma in the right upper arm within days of receiving the second dose of the Moderna vaccine, which may have been initially unrecognized until local symptoms brought attention to it [ 21 ], while another case revealed tumor regression after receiving the initial doses of the Moderna vaccine [ 22 ]. The variability presented in these cases raises the possibility of immunogenicity and reactogenicity varying based on an individual’s physiology and history of underlying oncologic and orthopedic conditions.…”

“…Regarding solid tumours that developed soon after receiving the modRNA COVID-19 vaccination, two cases were reported: one involved a 64-year-old woman who had a significant history of previously excised cutaneous melanoma reoccurring to the breast [52], and the other involved an aggressive sarcoma that developed at the injection site shortly after the second dose of Moderna [53].…”

“…In addition, the spike protein may interact with the tumor suppressor [ 77 , 78 ] (p. 53). It would therefore be wise to explore the tumorigenic effect in vivo and to monitor any cancers developed by vaccinated individuals over the long term, especially as it has been suggested that cancers can be reactivated by mRNA vaccines [ 79 , 80 , 81 ] or may develop after mRNA vaccination [ 82 , 83 , 84 , 85 , 86 ].…”
