For the purpose of broadening the available genetic resources to improve wheat breeding and to elucidate wheat evolution, 16 accessions of Aegilops tauschii newly collected in North Caucasia named NCT accessions were characterized genetically based on morphology, chloroplast SSR variation and AFLP. Ae. tauschii is one of the most important wild wheat genetic resources because it is the progenitor of the D genome of hexaploid wheat. Since Caucasia is considered to be a center of diversity of both cultivated and wild wheat, a lot of studies have been conducted to evaluate the diversity of Caucasian genetic resources including Ae. tauschii. Such kind of analyses, however, focused on Transcaucasia but little attention has been paid to North Caucasia because of the lack of available genetic resources. Based on the molecular analyses in this study, the 16 NCT accessions were generally divided into two groups although morphologically those are classified into the same subspecies. The grouping also represented geographical distribution, that is, the northern part group and Derbent group. This division is consistent with the two major genepools in Ae. tauschii reported in previous studies. The northern part and Derbent groups correspond to Eurasian wide genepool (called Tauschii genepool) and Caucasia and Caspian coast limited genepool (Strangulata genepool), respectively. Regarding to chloroplast, all the 16 accessions were genotyped as HG7, the most major haplogroup of the species. Although all the 16 NCT accessions were categorized into ssp. tauschii morphologically, accessions of Derbent group showed a tendency to have larger spikelets. Among at almost the largest level in ssp. tauschii. The results of this study filled the missing information of Ae. tauschii and will be helpful for future utilization.

An increase in cereal production is required if we are to support a world population of more than nine billion people expected in 2060. For this purpose, plant breeding will serve as a key technology as it did during the Green Revolution of the 1960s. However, the changing climate and decrease in agricultural resources are new challenges that will require consideration. We developed common and durum wheat populations expressing the intraspecifi c diversity of wild species. We collectively named these populations 'multiple derivative lines', and specifi cally 'multiple synthetic derivatives' (MSD) in the case of common wheat. The germplasm in the MSD population shows diverse morphology in regular genetic background enabling accurate selection of desired genotypes/phenotypes associated with abiotic stress tolerance. Accordingly, such plant materials can subsequently be used for various breeding purposes. Abiotic tolerance, especially drought tolerance, is largely determined by the interaction between genotype and environment, making drought-tolerance breeding diffi cult. Wild species, even the self-pollinating ones, possess high diversity which allows them to adapt to the diverse and fl uctuating environments. Accordingly, during wheat breeding for abiotic tolerance, it may be better to consider using diverse populations such as the MSD.

This work laid the foundation for cloning of shattering gene Br2 and provided first line of evidence that two major Aegilops tauschii lineages are differentiated by an inversion polymorphism. Chromosome inversions often accompany population differentiation and capture local adaptation during speciation. Aegilops tauschii, the D-genome donor species of hexaploid wheat, consists of two genetically isolated lineages, L1 and L2, but little is known about the genetic mechanisms underlying the population differentiation in this diploid species. During fine mapping of the shattering gene Br2 using a large F2 population derived from a cross between TA1604 (an L1 accession) and AL8/78 (an L2 accession), we found contrasting patterns of crossover distribution in the Br2 interval and neighboring regions despite the high local gene synteny with Brachypodium distachyon and rice. Br2 was localized in a 0.08-cM interval, and 13 marker loci formed a block, where single-crossovers were completely suppressed, but double-crossovers were enriched with a recombination rate of ~11 cM/Mb. In contrast, in a neighboring region no double-crossover was recovered, but single-crossover rate reached 24 cM/Mb, which is much higher than the genome-wide average. This result suggests a putative inversion polymorphism between the parental lines in the Br2 region. Genotyping using the markers from the Br2 region divided a collection of 55 randomly sampled A. tauschii accessions into two major groups, and they are largely genetically isolated. The two groups correspond to the L1 and L2 lineages based on their geographic distribution patterns. This provides first evidence that inversions may underlie the evolution of A. tauschii lineages. The presence of inter-lineage inversions may complicate map-based cloning in A. tauschii and transfer of useful traits to wheat.
