Development of colloidal gold immunochromatographic signal-amplifying system for ultrasensitive detection of Escherichia coli O157:H7 in milk

Abstract: 16A colloidal gold immunochromatographic signal-amplifying test strip with a sandwich 17 format was successfully developed to detect Escherichia coli O157:H7. The detection 18 signal was improved by addition of enhancers (HAuCl 4 and NH 2 OH·HCl) to the 19 nitrocellulose membrane of a conventional immunochromatographic strip. The 20 detection sensitivities of the test strip before and after signal amplification were 4×10 4 21 and 5×10 3 CFU mL -1 , respectively, as determined by a colloidal gold strip reader. … Show more

“…In all these cases, the decrease in LOD is due to higher coloration of the formed enlarged particles in comparison with the initial seed particles. For bacteria, the LOD decrease observed was in the range from 8 ( Escherichia coli O157: H7 in milk, [51]) to 100 times (for Salmonella enteritidis in milk [29]).…”

“…Ten grams each of non-C. sakazakii PIF sample was homogenized in 90 mL of nutrient broth medium and C. sakazakii (6 cfu), followed by incubation at 37°C and shaking at 160 rpm (Wang et al, 2015;Shukla et al, 2016). Aliquots of 1 mL from each bottle at 3, 4.5, 6, 7.5, and 9 h incubation periods were collected.…”

“…For comparison, the CG-ICA and MNP-ICA were developed and optimized according to previous work. 19,27 The procedure of CG-ICA and MNP-ICA were conducted as follows. A total of 8 mL of ICG or 8 mg of IMNPs was added in 100 mL of a series of S. enteritidis dilutions (1.95 Â 10 3 to 1.95 Â 10 8 CFU mL À1 ), and 100 mL of the mixture was transferred into the test strip.…”
