Development of chitosan–pullulan composite nanoparticles for nasal delivery of vaccines:in vivostudies

Abstract: Here, we aimed at developing chitosan/pullulan composite nanoparticles and testing their potential as novel systems for the nasal delivery of diphtheria toxoid (DT). All the chitosan derivatives [N-trimethyl (TMC), chloride and glutamate] and carboxymethyl pullulan (CMP) were synthesised and antigen-loaded composites were prepared by polyion complexation of chitosan and pullulan derivatives (particle size: 239-405 nm; surface charge: +18 and +27 mV). Their immunological effects after intranasal administration … Show more

“…Nanoparticles (NPs) of chitosan and trimethyl chitosan (TMC) are considered as one of the most attractive nasal delivery systems as it offers safety, biodegradability, biocompatibility, mucoadhesion, penetration enhancement and adjuvanticity [ 6 , 7 ]. Therefore, chitosan and TMC NPs have recently been used in the development of vaccines through nasal applications [ 8 12 ]. These NPs elicit strong immune response as shown by the significant increase in circulating antibody and the titer of secretory IgA in nasal washes [ 11 , 13 , 14 ].…”

“…44 PL hydrogel NPs have been employed for gene delivery 67 or as composite with CS for vaccine delivery. 68,69 Recently, PL was utilised as block in double hydrophilic block copolymers, which were utilised in turn for self-assembly, for example in the formation of completely hydrophilic capsules, 70 droplets 71,72 or particles. 73 Similar to DEX-based NPs, PL-based NPs (PLNPs) can be prepared by self-assembly after the hydrophobic modification of PL.…”
