Development of backcross recombinant inbred lines between Oryza sativa Nipponbare and O. rufipogon and QTL detection on drought tolerance

Abstract: Backcross populations at advanced generations are useful in evaluating the agronomic traits from unadapted germplasms, such as wild species. In this study, we generated 159 backcross recombinant inbred lines (BRILs) at the BC 2 F 8 generation between Oryza sativa Nipponbare (a recurrent parent) and O. rufipogon W630 (a donor parent). These BRILs were found to have nearly homozygous genome constitutions based on the marker genotypes at 180 SSR loci covering whole chromosomes. Since their selfed progenies have a… Show more

“…The wild accession was kindly provided by the National Institute of Genetics, Japan. A single wild plant of O. rufipogon W630 was crossed twice with O. sativa Nipponbare as previously described [17], and by the single-seed-descendant method, 159 BRILs were produced at the BC 2 F 8 generation [18]. These BRILs theoretically contain 12.5% of the wild genome in the genetic background of O. sativa Nipponbare.…”

“…The genotypes for 159 BRILs at 180 polymorphic simple sequence repeat (SSR) marker loci between Nipponbare and W630 were previously obtained [18]. With these marker data and the phenotypic data obtained in the present study, QTL analysis for eight floral and two grain traits was carried out by composite interval mapping using WinQTL Cartographer ver.…”

“…Estimation of wild chromosomal segments in each of the 159 BRILs at BC2F8 generation ranged between 0.0 to 23.6% (Thanh et al, 2011). The estimation was done using 180 microsatellite loci covering 1,362 cM of the 12 rice chromosomes.…”

“…These four species are known for its traits related to tolerance/resistance to biotic and abiotic stress (Thanh et al, 2011;Nonomura et al, 2010;Vaughan et al, 2005, Rangel et al, 2006. In the 2010/2011 growing season, when evaluating the F3 generation, segregant populations originated from crosses between O. glaberrima and SCS 112 presented good agronomic traits and a productivity of more that 10 t/ha.…”
