Partial-onset seizures are treated with Brivaracetam, which is a chemical butanamide derivative. Brivaracetam (BRV) in pharmaceutical and biological samples can be determined using various analytical techniques, including HPLC, UV-visible, and hybrid techniques including LC-MS, LC-MS/MS, UPLC-MS/MS, & other techniques, all of which are discussed in this review article. The review will analyze BRV determination using a variety of analytical approaches in comparison with each other. The findings of this review paper can be used as a starting point for future analysis of BRV.

Racetams are a class of medications defined by the presence of 2-pyrrolidone nucleus and are derivatives of gamma-aminobutyric acid (GABA), which acts as a major inhibitory neurotransmitter in the brain. Levetiracetam (second generation anti epileptic drug) and brivaracetam (third generation anti epileptic drug) are promising examples of prescribed antiepileptic racetams that act on SV2A. Brivaracetam is a newer antiseizure medication than levetiracetam. It has a more selective action on the synaptic vesicle glycoprotein 2A binding site, and it seems to provide a more favorable neuropsychiatric profile. Bioanalytical methods are widely used for quantitative estimation of drugs and their metabolites in biological matrices. Various analytical methods such as spectrophotometry, HPLC, GC, TLC, qNMR and UPLC have been used in laboratories for the quantitative analysis of antiepileptic racetams in biological samples throughout all phases of clinical research and quality control. The article incorporates various reported methods developed to help analyst in choosing crucial parameters for new method development of antiepileptic racetams and its derivatives and also enumerates metabolites, impurities reported so far.

It is clear from the literature that there are only a few methods reported with limitations of sophisticated instruments and poor sensitivity for the assay of Brivaracetam, and, particularly, no report on the simultaneous determination of the assay of Brivaracetam and its enantiomers (En) and diastereomers (DS) in a single method. Hence, it was our goal to build on a robust high-performance liquid chromatography (HPLC) process to the simultaneous assessment of Brivaracetam, and its En and DS, from the Brivaracetam drug. Accordingly, the reverse-phase chiral HPLC method development was established in a constructive way, such as buffer selection, column selection, mobile phase composition, organic modifier compositions, and column temperature variation. As a consequence, the simultaneous assessments were achieved in Daicel CHIRALPAK IG-3 (250 mm x 4.6 mm x 3 µm) with reverse-phase setup conditions consisting of 10 mM (NH 4 )HCO 3 buffer: CH 3 CN (1:1) %v/v in an isocratic elution. High sensitivity was obtained for Brivaracetam and its stereoisomers at Limit of quantitation (LOQ) 0.25 µg/ml and Limit of detection (LOD) 0.08 µg/ml. This method is systematically validated by the International Council for Harmonization procedures. Thus, the method is applicable in quality control, in drug analysis, and in the investigation of counterfeit drugs as well.
