The bisdichloroacetyldiamine WIN 18,446 reversibly inhibits spermatogenesis in many species, including humans; however, the mechanism by which WIN 18,446 functions is unknown. As retinoic acid is essential for spermatogenesis, we hypothesized that WIN 18,446 might inhibit retinoic acid biosynthesis from retinol (vitamin A) within the testes by inhibiting the enzyme aldehyde dehydrogenase 1a2 (ALDH1a2). We studied the effect of WIN 18,446 on ALDH1a2 enzyme activity in vitro, and on spermatogenesis and fertility in vivo, in mature male rabbits for 16 weeks. WIN 18,446 markedly inhibited ALDH1a2 enzyme activity in vitro with an IC50 of 0.3 μM. In vivo, the oral administration of 200 mg/kg WIN 18,446 to male rabbits for 16 weeks significantly reduced intratesticular concentrations of retinoic acid, severely impaired spermatogenesis, and caused infertility. Reduced concentrations of intratesticular retinoic acid were apparent after only 4 weeks of treatment and preceded the decrease in sperm counts and the loss of mature germ cells in tissue samples. Sperm counts and fertility recovered after treatment was discontinued. These findings demonstrate that bisdichloroacetyldiamines such as WIN 18,446 reversibly suppress spermatogenesis via inhibition of testicular retinoic acid biosynthesis by ALDH1a2. These findings suggest that ALDH1a2 is a promising target for the development of a reversible, nonhormonal male contraceptive.

BackgroundDevelopment of a non-hormonal long-acting reversible contraceptive for men could have a significant impact on reducing unintended pregnancies. Vasalgel™ is a high molecular weight polymer consisting of styrene-alt-maleic acid (SMA) dissolved in dimethyl sulfoxide being developed as a reversible male contraceptive device. It forms a hydrogel when implanted into the vasa deferentia, which prevents the passage of sperm. Previous studies in the rabbit have proven its efficacy, durability and rapid onset. This study evaluates the capacity to restore sperm concentrations in ejaculates after a reversal procedure.MethodsSodium bicarbonate was injected into the vasa deferentia after fourteen months of azoospermia following the injection of two device variations (Vasalgel 100 and Vasalgel 80). Semen samples were then collected for six months and sperm characteristics were compared to baseline levels. Samples of vasa deferentia were obtained for histological examination.ResultsSpermatozoa were present in all subject ejaculates after the reversal procedure. Sperm concentration and sperm motility were similar to baseline levels after reversal, while sperm forward progression was significantly lower and normal acrosomes were not observed. Forward progression percentages increased linearly during six months of semen collection, however, normal acrosomes were not observed at the conclusion of the study. Histologically, several vasa deferentia were clear of the device and contained an intact epithelial lining. A smaller proportion of tissues contained residual test material. A secondary intraluminal inflammatory response was seen occasionally in the tissues containing residual material. There was no difference between the two device variations for studied parameters.ConclusionsVasalgel’s prevention of sperm transport for 14 months was reversed through an intravasal injection of sodium bicarbonate. Post-reversal sperm concentrations and motility returned to baseline levels during the six-month follow up. Residual material in the vas lumen or compromised epididymal and vas deferens function may be resulting in reduced forward progression and loss of acrosomes during transit through the vas. Reduced forward progression and the lack of normal acrosomes strongly suggest impaired sperm function.

This study was designed to evaluate mature New-Zealand White (NZW)male rabbits physiological and reproductively as affected by different levels of adenosine as growth hormone on reproductive functions of male rabbits. Eighteen white New Zealand mature male rabbits aged 5-6 months and weighing (2.7-2.9 kg) were divided into three groups (6 rabbits each). First group of rabbits served as a control group, second and third rabbit groups were injected intravenously by (0.4 and 0.8 mg of adenosine/ animal/48hours) respectively, dissolved in 0.5 ml (0.9 NaCl) for 60 days. Blood samples were taken once to evaluate blood components and semen was collected twice to evaluate physical semen characteristics from each animal every 10 days. Best results were recorded in the second group of animals (injected with 0.4 mg adenosine/animal/48 hours), where the mean was (7.8gm/dL) for total serum protein, (14.5gm/dL) for hemoglobin, (9.8gm/dL)for calcium concentration and (7.05nmol/L) for testosterone hormone level that increased significantly (P<0.05) compared to other groups at the end of the experiment. Also, the results the second group animals presented (0.97 ml) for semen ejaculating volume, (85%) for sperm motility, (91.8%) for sperm live that significantly increased at (P<0.05) compared to other groups and showed a significant reduction of (6%) for sperm abnormalities than those found in other groups of animals at 60 days of experimental time. Conclusively, this study shows that the injection by 0.4 mg adenosine/animal every 48 hours for 60 days continuously improved physical semen characteristics, blood components and can safely be used on rabbit males.
