Development of an Inexpensive Artificial Vagina for Semen Collection from Rabbits

Abstract: The rabbit is well suited for infertility research because of the animal's size, relatively low cost, and accessibility of the genitalia. There are several studies reporting sperm count and motility in rabbits; however, no easily reproducible or inexpensive device for semen collection has been reported. Herein, we report the construction of an inexpensive, effective artificial vagina assembled from easily obtainable products that may be used to collect rabbit ejaculates. This device was used to perform 243 eja… Show more

“…The WIN 18,446 was compounded in a simple syrup base at a concentration of 100 mg/mL with banana crème flavoring and dosed orally using a syringe. Spermatogenesis was assessed by having the animals ejaculate sperm into an artificial vagina at baseline and weekly during treatment and recovery (Naughton et al, 2003). Sperm count, concentration, and motility were quantified manually using a hemocytometer (Seed et al, 1996).…”

“…Semen collections were performed using a warmed artificial vagina semen-collection device designed for use with the rabbit and a “teaser” female to encourage mounting [11]. Semen specimens were evaluated for volume, total sperm count, motility and forward progression using manual methods.…”

“…Semen samples were collected from each group after ten days of treatment and every 10 days, till the end of the experiment with a rabbit artificial vagina, (two ejaculates per male, twice every 10 days) using thermal and mechanical stimulation to stimulate ejaculation. Procedure of the collection was performed according to (Naughton et al, 2003).Physical semen characteristics as semen volume, sperm motility, sperm live and sperm abnormality were assessed for the semen sample, similarly to those described by (Essig et al, 2007 andQuinteiro et al, 2007).…”
