Development of an enzyme immunoassay for the serodiagnostic of tuberculosis and mycobacterioses

Abstract: Abstract. An enzyme-linked immunosorbent assay (ELISA) test has been developed for the serodiagnosis of mycobacterioses. The test is based on an interspecific antigen, antigen 60 (A60). This antigen belongs to the group of thermostable macromolecular antigens and is the principal constituent of tuberculin RT 23. A60 is composed of proteins, polysaccharides and lipids in a ratio that varies with the growth cycle of the mycobacterium. The test detects antibodies against mycobacteria and is most effective in case… Show more

“…In particu lar. the evaluation of IgG directed against the antigen 60 (A60) [12][13][14] -the main thermostable component of old tuberculin and purified protein derivative -provided high diagnostic relevance [15][16][17][18][19], On the other hand, dis cordant results were reported on the diagnostic accuracy of evaluation of the IgM-mcdiated humoral immune response [20,21], In this work anti-A60 IgA levels in patients with PTb were measured by ELISA and compared to the anti-A60 IgG titers.…”

“…The interferon test is a costly substitute to a much cheaper method of assessment of latent infections, by monitoring the humoral immunity. The Anda TB serological test is available since 1989 [58] was shown useful for the detection of latent infections in exposed groups in 1989 [59] and 1990 [60], among some cancer patients in 1995 [61]. Serological measurements also give valuable prognostic and diagnostic clues in BCG vaccinated infants [22].…”

“…The hummoral immune response following BCG vaccination indicates that IgM anti PPD increase progressively until the third month, followed by an increase of IgG. The antibody dynamics in TB infection could be similar (Maes et al, 1989), but it also depends on the clinical form and treatment starting. IgA serum antibodies appear immediately after IgM and precede IgG, but the CSF dynamics could vary.…”
