Development of an ELISA to detect clenbuterol in swine products using a new approach for hapten design

Abstract: This research outlines the application of an enzyme-linked immunosorbent assay (ELISA) for the analysis of clenbuterol in animal products. Our assay showed good sensitivity for clenbuterol (0.4 ng/g or 0.4 ppb) and low detection limit (0.09 ng/g or 0.09 ppb). A low cross-reactivity for other β2-agonist drugs such as salbutamol, terbutaline, and epinephrine led to formatting an ELISA kit considered to have a high specificity for clenbuterol. A survey of Ho Chi Minh City pork market was conducted as part of the … Show more

“…Especially, the characteristic structural activity of the hapten may have huge effects on it. Although the significance of spacer arm properties (length, flexibility, and rigidity) has been discussed in many studies, there remains a lack of in-depth exploration of a specific relationship between the structural activity of hapten and their immunoinducing ability. Hence, we attempted to infer hapten structure that is more favorable for inducing antibody production based on their structural information (e.g., three-dimensional conformation, size, charge distribution, antibody–antigen recognition).…”

“…Clenbuterol [35,36], salbutamol [37] and phenylethanolamine A [38] in various samples (animal urine, blood, hair and tissues) have been measured using direct competitive ELISA. The mAb or pAb against β 2 -agonist is coated onto the microtiter plate, the β-agonist in sample and β 2 -agonist labeled with HRP or ALP competes for binding with the mobilized antibody.…”

“…There are many ways to detect CB, including high performance liquid chromatography (HPLC) [ 8 ], capillary electrophoresis (CE) [ 9 ], enzyme-linked immunosorbent assay (ELISA) [ 10 , 11 ], gas chromatography-mass spectrometry (GC-MS) [ 12 ], and liquid chromatography with tandem mass spectrometry (LC-MS) [ 13 , 14 ]. Although these detection methods have the advantage of high accuracy, professional operators, complex extraction steps, and expensive equipment are also needed.…”
