Nitrite plays a critical role in a variety of physiological processes and maintaining the nitrite level in an appropriate range is vital to keep healthy. Current nitrite analysis methods lack sensitivity and require tedious operations, which could not meet the need of point-of-care (POC) nitrite detection in precision medicine. Here we present a cyanocobalamin (VB12) bionic enzyme-assisted ion-selective amperometric biosensor based on 3D porous conductive matrix (PCM), which can facilitate rapid and accurate POC nitrite monitoring in complex biofluids. The experimental findings quantitatively demonstrate that the biosensor has a sensitivity of 64.08 μA/(mM·cm2), a wide linear range of 0.025–45 mM, and low limit of detection of 1 nM. Moreover, the developed VB12/BSA-PCM biosensor shows outstanding stability after 21 days with 2% decline in current signal, and high repeatability between batches with RSD of only 1.29%. Real salivary nitrite detection has been evaluated, and the results match well with the commercial nitrite analyzer. Thus, the bionic enzyme-assisted ion-selective amperometric biosensor proposed herein has potential utility as an affordable tool for POC detection and home-based healthcare.

This study presents a rapid microfluidic paper-based analytical device (μPAD) capable of simultaneously monitoring Gram-negative bacteria and nitrite ions (NO2 –) for water quality monitoring. We utilize gold nanoparticles (AuNPs) functionalized with polymyxin molecules (AuNPs@polymyxin) to cause color change due to aggregation for the detection of Gram-negative bacteria, and antiaggregation in the presence of o-phenylenediamine (OPD) for NO2 – detection. In this study, Escherichia coli (E. coli) serves as the model of a Gram-negative bacterium. Using the developed μPADs, the color changes resulting from aggregation and antiaggregation reactions are measured using a smartphone application. The linear detection ranges from 5.0 × 102 to 5.0 × 105 CFU/mL (R 2 = 0.9961) for E. coli and 0.20 to 2.0 μmol/L (R 2 = 0.995) for NO2 –. The detection limits were determined as 2.0 × 102 CFU/mL for E. coli and 0.18 μmol/L for NO2 –. Notably, the newly developed assay exhibited high selectivity with no interference from Gram-positive bacteria. Additionally, we obtained acceptable recovery for monitoring E. coli and NO2 – in drinking water samples with no significant difference between our method and a commercial assay by t test validation. The sensor was also employed for assessing the quality of the pond and environmental water source. Notably, this approach can also be applied to human urine samples with satisfactory accuracy. Furthermore, the assay’s stability is extended due to its reliance on AuNPs rather than reagents like antibodies and enzymes, reducing costs and ensuring long-term viability. Our cost-effective μPADs therefore provide a real-time analysis of both contaminants, making them suitable for assessing water quality in resource-limited settings.

Recirculating aquaponics integrate aquaculture and hydroponics with the aid of microorganisms to ensure a sustainable supply of fish and vegetables. In this study, we designed and constructed a flood-and-drain aquaponic system with Clarias gariepinus as the aquaculture and Celosia argentea, Corchorus olitorius, and Ocimum gratissimum as plant components. Nitrogenous waste from the aquaculture unit was circulated to the plant growth beds, which served as bio-oxidizers of toxic ammonia to nitrate and returned less toxic water back to the aquaculture unit. An evaluation of the growth parameters of C. gariepinus in aquaponics and control fish tanks revealed that aquaponics-raised C. gariepinus gained an additional 205.6% of their initial mean weight, whereas those grown in the control fish tank gained an additional 182.2% of their initial mean weight. The majority (37.5%) of aquaponics-raised C. gariepinus weighed 750 g -1 kg, whereas the highest percentage of C. gariepinus raised in the control fish tank (23.4%) weighed 500 g -700 g. C. gariepinus raised in aquaponics had significantly higher gross feed conversion efficiency and protein efficiency ratio than those raised in the control tank. The mortality rate in the aquaponic fish tank was 0% compared to the 11.43% mortality rate in the control fish tank. There was no significant difference in the nutritional composition of C. gariepinus raised in either tank; however, the aquaponic fish tank had a higher nitrogen retention rate. This study showed that C. gariepinus raised in aquaponics had better biomass accumulation than those raised in conventional fishponds.
