Development of a reverse transcription quantitative polymerase chain reaction-based assay for broad coverage detection of African and Asian Zika virus lineages

Abstract: The Zika virus (ZIKV) is an arbovirus that has spread rapidly worldwide within recent times. There is accumulating evidence that associates ZIKV infections with Guillain-Barré Syndrome (GBS) and microcephaly in humans. The ZIKV is genetically diverse and can be separated into Asian and African lineages. A rapid, sensitive, and specific assay is needed for the detection of ZIKV across various pandemic regions. So far, the available primers and probes do not cover the genetic diversity and geographic distributio… Show more

“…Viral stock titration by 50% tissue culture infective dose (TCID 50 ) TCID 50 was measured as previously reported [50]. In brief, Vero cells in 96-well plates were grown to 80% con uence and infected with 10-fold serial dilutions of the stock SARS-CoV-2 (BetaCoV/Shenzhen/SZTH-003/2020, GISAID No.…”

“…A quantitative RT-PCR targeting E gene has been very recently developed that could detect all Asian and African lineages of ZIKV. This assay alleviates the disadvantage of earlier developed real-time PCR as they could not detect all strains of ZIKV and, moreover, this assay also showed a detection limit of 5 RNA transcript copies (Yang et al, 2017 ). A list of ZIKV detection assays is presented in Table 1 .…”
