Development of a Phage Display Panning Strategy Utilizing Crude Antigens: Isolation of MERS-CoV Nucleoprotein human antibodies

Abstract: Antibody phage display has been pivotal in the quest to generate human monoclonal antibodies for biomedical and research applications. Target antigen preparation is a main bottleneck associated with the panning process. This includes production complexity, downstream purification, quality and yield. In many instances, purified antigens are preferred for panning but this may not be possible for certain difficult target antigens. Here, we describe an improved procedure of affinity selection against crude or non-… Show more

“…A large phage-displayed human naive scFv library (Mehta I/II) with 2.7 × 10 10 clones from the Dana-Farber Cancer Institute was used as a resource for the isolation of human antibodies against MERS-CoV [135]. In another project, a research group in Malaysia improved panning strategies with a naïve human scFv library (library size of 10 9 ) to successfully identify mAbs specific to the MERS-CoV nucleoprotein [136].…”

“…Phage display technique of producing antibodies was developed for MERS-CoV and effectively applied in light of the fact that bacteriophages have the potential to produce recombinant antibodies (Ab) rapidly [12]. Another Yin-Yang biopanning technique features the chance of utilizing crude antigens for the isolation of monoclonal Ab by phage display method [13]. Before using these expensive techniques, production of artificial Ab was primarily done by using animals but it is a slow process and less cost effective than using bacteriophage display techniques [14].…”

“…They successfully displayed antibody variable regions on phages by using immunoglobulin variable genes of hybridomas and B cells. Since then, phage display technology has been extensively used for the discovery of antibodies or peptides against a large variety of antigens in many fields of application such as toxiconology (Ledsgaard et al, 2018), drug discovery (Erdag et al, 2007;Mimmi et al, 2019), immunization (Bahadır et al, 2011), epitope mapping (Folgori et al, 1994) and virus or toxin neutralization (Lim et al, 2019) by using phage peptide and antibody libraries.…”
