A monoclonal antibody-based flow-through immunoassay (FTA) was developed using a nitrocellulose membrane placed on the top of adsorbent pads enclosed in a plastic cassette with a test zone at the center. The FTA could be completed within 10 min. Clear purple dots against a white background indicated the presence of Aphanomyces (A.) invadans. The FTA limit of detection was 7 µg/mL for A. invadans compared to 56 µg/mL for the immunodot. FTA and polymerase chain reaction (PCR) could detect A. invadans in fish tissue homogenates at a 10-11 dilution compared to a 10-8 dilution by immunodot. In fish suffering from natural cases of epizootic ulcerative syndrome (EUS) collected from Mangalore, India, FTA and PCR could detect A. invadans in 100% of the samples compared to 89.04% detected by immunodot. FTA reagents were stable and produced expected results for 4 months when stored at 4~8℃. This rapid test could serve as simple and cost-effective on-site screening tool to detect A. invadans in fish from EUS outbreak areas and in ports during the shipment of live or frozen fish.

Grouper iridovirus (GIV) belongs to the Ranavirus genus and is one of the most important viral pathogens in grouper, particularly at the fry and fingerling stages. In this study, we identified and characterized the GIV-2L gene, which encodes a protein of unknown function. GIV-2L is 1242 bp in length, with a predicted protein mass of 46.2 kDa. It displayed significant identity only with members of the Ranavirus and Iridovirus genera. We produced mouse monoclonal antibodies against the GIV-2L protein by immunizing mice with GIV-2L-His-tag recombinant protein. By inhibiting de novo protein and DNA synthesis in GIV-infected cells, we showed that GIV-2L was a late gene during the viral replication. Finally, immunofluorescence microscopy revealed that GIV-2L protein accumulated in both the nucleus and cytoplasm of infected cells. These results offer important insights into the pathogenesis of GIV.

White spot syndrome virus (WSSV) has been extensively investigated since the white spot disease outbreak in shrimp in 1990. These investigations have mainly focused on determining the function of viral structural proteins, and on utilizing the envelope proteins as subunit vaccines to protect the host. These studies have shown that a WSSV vaccine can be a practical and effective approach for controlling WSSV infection. The development of such a vaccine has become an intense focus of research in the field. This research has resulted in significant progress in the development of WSSV vaccines. However, because of the progress of this work, periodic summaries are needed to facilitate further research on the development of effective WSSV vaccines. This paper provides a comprehensive summary on the status of WSSV vaccines with regard to the following aspects: subunit vaccines, inactivated whole virus vaccines, DNA vaccines, protective antibodies, and the application of RNAi technology. Current limitations in this area of research are also described, as well as prospects for the development and application of improved WSSV vaccines in the future, and for investigating other important aquatic diseases.
