Development of a microtitre plate enzyme immunoassay for the determination of progesterone

Abstract: A rapid, solid-phase microtitre plate enzyme immunoassay (EIA) for progesterone is described using progesterone 3-O-carboxymethyloxime-horseradish peroxidase as the label and an antiserum raised in rabbits to a progesterone 11 alpha-hemisuccinyl-bovine serum albumin immunogen. A competitive reaction was used with a reaction time of 2 h. Antibody-bound and free steroid were separated in a simple washing step of the antibody-adsorbed well surface. 2,2'-Azino-di-(3-ethylbenzthiazoline sulphonic acid) diammonium s… Show more

“…Blood samples were chilled on ice and centrifuged at 2,100 rpm for 15 min at 4 C. The plasma was aspirated without disturbing the buffy coat and frozen until needed. Plasma progesterone levels were determined by a rapid solid phase microtiter plate enzyme immunoassay (EIA), 24 using progesterone 3-0-carboxymethyloime-horseradish peroxidase as the label and antiserum raised in rabbits to a progesterone 11 alpha-hemisuccinyl bovine serum albumin immunogen. A competitive reaction was used with a reaction time of 2 hr.…”

“…Cortisol plasma levels were determined using an enzyme immunoassay (EIA). The technique was performed following the protocol developed by Munro and Stabenfeldt (1984) and modified by the Wisconsin Regional Primate Research CTR. Intra-and inter-assay coefficients of variation were 4.5 and 16.9% for low pool and 3.1 and 13.8% for high pool, respectively.…”

“…Cats that were before ovulation at insemination received the second administration immediately after insemination, while those after ovulation at insemination did not receive the second administration. To evaluate the success of induction of ovulation, the peripheral progesterone (P) level was measured 15 days after hCG administration by EIA [6] in cats that were pre-ovulatory at AI excluding pregnant animals, and when the P level was increased, induction of ovulation was judged successful.…”
