Development of a mathematical model for evaluating the dynamics of normal and apoptotic Chinese hamster ovary cells

Abstract: A metabolic flux based methodology was developed for modeling the metabolism of a Chinese hamster ovary cell line. The elimination of insignificant fluxes resulted in a simplified metabolic network which was the basis for modeling the significant metabolites. Employing kinetic rate expressions for growing and non-growing subpopulations, a logistic model was developed for cell growth and dynamic models were formulated to describe culture composition and monoclonal antibody (MAb) secretion. The model was validat… Show more

“…Specifically, the growth functions ( r i ) are glutamate-dependent (Equation 7). This dependency was introduced since it has been reported that glutamine, or glutamate in the GS system [50], is the main energy-yielding metabolic route [11, 51, 52] with over 90% flowing into the TCA cycle [53]. In addition, the cyclin growth functions ( r i = G1,G2 ) were also formulated as lactate-dependent since it has been reported [54, 55] that its concentration inhibits cell growth.…”

“…In recent years, several studies have appeared where stationary MFA was combined with a kinetic model to describe metabolic flux dynamics in batch and fed-batch cultures of CHO cells [35,107,108,[110][111][112][113]. The initial kinetic modeling framework was based a lumped network model derived from the CHO cell model by Provost and Bastin [110].…”
