Development of a Homogeneous Assay for Measurement of Small Dense LDL Cholesterol

Abstract: BACKGROUND: Plasma concentrations of small dense (sd)-LDL are associated with the prevalence of cardiovascular events. However, the special equipment and long assay times required for sd-LDL measurement have hindered its clinical application. Herein, we report development of a simple homogeneous assay for sd-LDL-cholesterol (C) adaptable to autoanalyzers.

“…Serum glucose was also measured. For the purposes of this study, we used archived plasma samples that had been frozen at −80 ℃ and never previously thawed for the assessment of direct LDL-C and sd-LDL-C by homogeneous methods on a Hitachi 7180 automated analyzer (Hitachi, Tokyo, Japan) 18,19) . The kits used for these tests (LDL-C and sd-LDL-C) were provided by Denka Seiken (Tokyo, Japan).…”

“…LDL-C and HDL-C were measured by the homogenous direct assay (LDL-EX and HDL-EX, Denka Seiken, Tokyo, Japan). Small dense LDL-C was directly measured by a homogenous method recently established [13]. In this manuscript, the value for HbA1c(%) is estimated as an NGSP(National Glycohemoglobin Standardization Program) equivalent value(%),as calculated by the formula HbA1c (%) = HbA1c (Japan Diabetes Society [JDS]) (%) +0.4%, considering the relational expression of HbA1c (JDS) (%) measured by the previous Japanese standard substance and measurement methods and HbA1c (NGSP) [14].…”

“…HDL cholesterol concentrations were measured using similar methods after precipitation of apolipoprotein B-containing lipoproteins with sodium phosphotungstate in the presence of magnesium chloride 21) . Plasma concentrations of small dense LDL cholesterol were measured by the simple homogeneous assay described by Ito et al 22) (sd-LDL-Ex SEIKEN, Denka Seiken Co. Ltd, Tokyo, Japan). The physicochemical principle of the method is as follows.…”
