Development of a Drug Delivery System Based on Chitosan Nanoparticles for Oral Administration of Interferon-α

Abstract: Despite the good clinical efficacy of interferon-alpha (IFNα) to treat some types of cancer and viral infections, this biological drug is underused given its severe adverse effects and high dosing parenteral regimens. Aiming to achieve a breakthrough in therapy with IFNα, this work reports for the first time on the design and full characterization of a novel nanomedicine of IFNα-2b-loaded chitosan nanoparticles (IFN-CT NPs) for oral delivery. IFN-CT NPs produced by ionotropic gelation, encapsulating approximat… Show more

“…Other encapsulation techniques, such as electrospraying, are known to obtain a narrow particle size distribution; this is not always the case with IG. Polydispersion indices (PDI) are usually higher than 0.1, reaching values up to 0.5 . This indicates a heterogeneity in particle size that not only could affect the quantity of the drug loaded but could also limit the interaction of the formulation with biological structures.…”

“…The study was aimed to assess the possible application of this system for bone tissue engineering. Similarly, Biglione et.al., described the design and full characterization of a novel CS based nano carrier for the oral delivery of IFNα‐2b protein that is used to treat different types of cancer and viral infections, aiming to replace its parenteral administration.…”

“…Oral administration is the best therapeutic way for intestinal diseases because it is regarded as the simplest and fastest method for drugs to reach the intestine and contact with the target site directly. [ 8 ] To meet the demand for oral administration, the oral drugs for intestinal protection should hold the following characteristics: i) Good stability and protective ability remain unchanged in the gastric juice environment. ii) Long residence time for drugs in the intestine is required for its high utilization.…”
