Solubility Measurement and Various Solubility Parameters of Glipizide in Different Neat Solvents

Glipizide (GLZ) is an oral hypoglycemic agent, which is a weakly aqueous soluble drug. The solubility values of GLZ in various neat solvents are scarce in the literature. Hence, the solubility of GLZ in 12 different neat solvents, namely, "water, methanol, ethanol, isopropanol (IPA), 1-butanol, 2-butanol, ethylene glycol (EG), propylene glycol (PG), poly(ethylene glycol)-400 (PEG-400), ethyl acetate (EA), dimethyl sulfoxide (DMSO), and Transcutol-HP (THP)", at "T = 298.2−318.2 K" and "p = 0.1 MPa" was measured. The recorded solubilities of GLZ were correlated by "van't Hoff and Apelblat models" using rootmean-square deviation (RMSD). The overall RMSD was obtained as 1.21 and 1.40% for "Apelblat and van't Hoff models", respectively. Different solubility parameters of all studied materials including drug and solvent were calculated to find the best solvent for GLZ. The solubilities of GLZ (expressed in mole fraction) have been found highest in DMSO (2.81 × 10 −2 ), followed by THP, EA, 2-butanol, 1-butanol, IPA, PEG-400, ethanol, PG, methanol, EG, and water (1.98 × 10 −4 ) at "T = 318.2 K". All investigated solubility parameters of GLZ were recorded very close to the DMSO. "Apparent thermodynamic analysis" showed an "endothermic and entropy-driven dissolution" of GLZ in the 12 different neat solvents. The highest molecular interactions were recorded in GLZ−DMSO compared to other combinations. Overall, DMSO has been considered as the best solvent for the solubilization of GLZ.

Intranasal Formulation and Characterization of Chitosan Microsphere for Improving in Vitro Mucoadhesion, Residence Time and Absorption Rate of Pregabalin

Objective: Chitosan-based pregabalin microsphere (CBPM) formulation was prepared to improve in vitro mucoadhesion and absorption of pregabalin via intranasal administration. Methods: The CBPM formulations were prepared using the inotropic gelation method and optimized using the Box-behnken design. The optimized CBPM formulation was physico-chemically characterized using scanning electron microscopy, thermal analysis, Fourier transform infrared spectrometry and powder x-ray diffraction. Additionally, the CBPM formulation was characterized for functional parameters such as in vitro mucoadhesion, in vitro drug release, ex vivo permeability across the sheep nasal mucosa and in vivo anticonvulsant activity in pentylenetetrazol (PTZ)-induced seizures model in mice. Results: The design-optimized CBPM exhibited a 91.45 % inclusion efficiency of pregabalin in the microspheres. The Physico-chemical analysis of the individual components and the optimized formulation confirmed the formation of CBPM. The in vitro mucoadhesion study revealed ~80% mucoadhesive of the CBPM to the sheep nasal mucosa. The in vitro dissolution profiles of CBPM was significantly higher (~97%) than that of pure pregabalin (~70%). The CBPM displayed a higher rate and extent of permeability (~90%) than pure pregabalin (~76%) across the sheep nasal mucosa. The in vivo anticonvulsant activity showed that intranasal administration of CBPM resulted in significant (P<0.01) protection against PTZ-induced convulsions in mice. Conclusion: The chitosan-based microsphere intranasal formulation could be employed as promising delivery for rapid pregabalin absorption.

Aim: Due to the presence of rich blood circulation and non-invasive nature of administration, the nasal route has been established as valuable therapeutic alternatives. This study was designed with the basic objective to formulate mucoadhesive microspheres of anticonvulsant drug, lamotrigine (LT) for intranasal delivery and to carry out its pharmacodynamic investigations to benefit the emergency cases of epileptic seizures. Materials and Methods:The microspheres were prepared by ionotropic gelation method and characterized for morphology scanning electron microscopy, particle size, drug entrapment efficiency, thermal behavior (differential scanning calorimetry), and crystallinity (X-ray diffraction), in vitro swelling studies, in vitro drug diffusion, ex vivo bioadhesion, and ex vivo biocompatibility studies in excised sheep nasal mucosa. The resultant LT loaded chitosan (CH) microspheres were further evaluated for pharmacodynamic efficacy in pentylenetetrazol (PTZ) induced seizures in mice. Result and Discussion: Results demonstrated that the microspheres were discrete, smooth, and spherical in shape with size 24.5 ± 2.62 to 48.52 ± 2.34 µm, appropriate for nasal drug delivery. The CH-based nasal LT microspheres demonstrated high encapsulation efficacy, strong bioadhesion potential, and high permeation without any signs of morphological toxicity in excised sheep nasal mucosa. Importantly the intranasal administration of LT microspheres delayed the onset of clonic convulsion and offered complete protection against the PTZ induced seizures in mice compared to its peripheral administration. Conclusion: Thus, the formulation of LT loaded CH mucoadhesive microspheres offers promising advantages over conventional dosage with its immediate onset of action.
