Development and validation of four one-step real-time RT-LAMP assays for specific detection of each dengue virus serotype

Abstract: Background4 one-step, real-time, reverse transcription loop-mediated isothermal amplification (RT-LAMP) assays were developed for the detection of dengue virus (DENV) serotypes by considering 2,056 full genome DENV sequences. DENV1 and DENV2 RT-LAMP assays were validated with 31 blood and 11 serum samples from Tanzania, Senegal, Sudan and Mauritania. DENV3 and DENV4 RT-LAMP assays were validated with 25 serum samples from CambodiaMethodology/Principal findings4 final reaction primer mixes were obtained by usin… Show more

“…Some highlights from adaptations of LAMP include multiplexed LAMP, electrochemical LAMP, and disc-based LAMP [46][47][48]. For RNA viruses, such as ZIKV, DENV, and CHIKV, it is necessary to perform a reverse transcription reaction LAMP (RT-LAMP) [27,39,42,[49][50][51][52] which includes the enzymes that first convert RNA → DNA upstream of the LAMP process, unless a LAMP enzyme with both reverse transcriptase and DNA polymerase is used [53].…”

“…Loop-mediated amplification (LAMP) is a modified version of PCR in which a uniform temperature ranging from 60 to 65°C is applied using a water bath for nucleic acid amplification and, thus, obviates the requirement of a thermocycler [39,40]. This approach to nucleic acid amplification relies on strand displacement reaction that produces a self-complementary singlestranded loop structure for binding the primers and amplifies target with high specificity and rapidity [41,42].…”

“…The developed RT-LAMP is found to be selective and did not cross-react with other types of Flavivirus. The assay for DENV1 and DENV2 were validated using blood and serum samples obtained from different regions [40].…”

“…Recent studies report the development of real-time LAMP assays using SYBR green for real-time detection, rather than using visual color change, interestingly, amplifying the targets ( P. falciparum ) from directly boiled blood samples [ 51 ]. To this direction of real-time LAMP, one step has been further accomplished to detect other non-malaria fever-related tropical infections, such as dengue [ 52 ] and chikungunya [ 53 ], using reverse transcriptase real-time (RT)-LAMP.…”
