This paper proposes a novel idea to enhance the sensitivity and selectivity of surface plasmon resonance (SPR) optical sensor for detection of dengue virus type-2 envelope proteins (DENV-2 E-proteins) using polyamidoamine (PAMAM) dendrimer biopolymer-based nanocomposite thin film. For this purpose, two ranges of DENV-2 E-protein concentrations, i.e., 0.000008–0.0001 nM and 0.00008–0.005 nM were evaluated, and the lowest detectable concentration was achieved at 0.00008 nM. The incorporation of PAMAM dendrimer-based nanocomposite thin film with an SPR sensor exhibited a significant increase in sensitivity and binding affinity to a lower range DENV-2 E-protein concentrations. Moreover, the proposed sensor displayed good selectivity towards DENV-2 E-proteins and have an average recovery of 80–120%. The findings of this study demonstrated that PAMAM dendrimer-based nanocomposite thin film combined with SPR sensor is a promising diagnostic tool for sensitive and selective detection of DENV-2 E-proteins.

Growth hormone ( GH ) is required for normal postnatal development in poultry; however, no immunoassay exists to assess its levels in geese plasma, hindering the study of endocrine regulation in this species. We developed a sandwich ELISA to determine the GH concentrations in the plasma of geese. Recombinant goose GH was produced using a eukaryotic expression system and purified for use as the reference standard in ELISA and the antigen for producing the polyclonal antibodies in rabbits. Rabbit anti-goose GH polyclonal antibody was used to coat the wells of the ELISA plate, and its biotinylated form served as the detection antibody. An avidin-conjugated horseradish peroxidase was used to bind the detection antibody and catalyze the chromogenic reaction of 3,3,5,5-tetramethylbenzidine and H 2 O 2 . A sigmoidal curve was fitted to the optical density and the log of the standard GH concentration using the four-parameter logistic model. The sensitivity of the assay was less than 0.156 ng/mL. The intra- and interassay coefficients of variation were less than 9 and 13%, respectively. The response curve of the serially diluted plasma samples from geese exhibited a good parallel relationship with that observed for the reference standards. The assay effectively detected differences in GH concentrations in plasma samples from geese at various physiological stages; thus, it will be useful for future study of their growth and metabolism.

The acute respiratory distress syndrome (ARDS) describes a heterogenous population of patients with acute severe respiratory failure. However, contemporary advances have begun to identify distinct sub‐phenotypes that exist within its broader envelope. These sub‐phenotypes have varied outcomes and respond differently to several previously studied interventions. A more precise understanding of their pathobiology and an ability to prospectively identify them, may allow for the development of precision therapies in ARDS. Historically, animal models have played a key role in translational research, although few studies have so far assessed either the ability of animal models to replicate these sub‐phenotypes or investigated the presence of sub‐phenotypes within animal models. Here, in three ovine models of ARDS, using combinations of oleic acid and intravenous, or intratracheal lipopolysaccharide, we investigated the presence of sub‐phenotypes which qualitatively resemble those found in clinical cohorts. Principal Component Analysis and partitional clustering identified two clusters, differentiated by markers of shock, inflammation, and lung injury. This study provides a first exploration of ARDS phenotypes in preclinical models and suggests a methodology for investigating this phenomenon in future studies.
