Development and validation of a high resolving power absolute quantitative per‐ and polyfluoroalkyl substances method incorporating Skyline data processing

Abstract: Rationale The ability to perform absolute quantitation and non‐targeted analysis on a single mass spectrometry instrument would be advantageous to many researchers studying per‐ and polyfluoroalkyl substances (PFAS). High‐resolution accurate mass (HRAM) instrumentation (typically deployed for non‐targeted work) carries several advantages over traditional triple quadrupole workflows when performing absolute quantitation. Processing this data using a vendor‐neutral software would promote collaboration for these … Show more

“…We used LC–HRMS techniques to check for nitrosamine contamination, showing how modern methods meet regulatory requirements. Jeffrey et al used Orbitrap Exploris 240 to find per‐ and polyfluoroalkyl substances, demonstrating the instrument's sensitivity with a lowest detection limit of 0.32 ng/L 40 . One limitation of our study is not using an internal standard.…”

“…The samples were randomized again and then analyzed with an Orbitrap Exploris 240 (Thermo Scientific, Bremen, Germany) incorporating a Thermo Scientific Vanquish Horizon LC system (Germering, Germany) using a previously published method [ 32 ]. Briefly, a 100 µL aliquot of each sample was injected onto a Kinetex F5 (2.1 × 100 mm, 100 Å; Phenomenex, Torrance, CA, USA) analytical column at 45 °C for separation.…”

“…PFMOAA interestingly had the largest peak area compared to the control when dried with heat. It should be noted that this analyte experiences sub-optimal reproducibility with most forms of LC, 34 and the replicate data at lower concentrations had a very high standard deviation. In general, this class of compounds has better recovery when dried without heat.…”
