Development and validation of a reliable method for studying the distribution pattern for opiates metabolites in brain

Abstract: Brain distribution pattern of "street" heroin metabolites (morphine and codeine) was investigated in two fatalities due to "acute narcotism". A suitable sample pretreatment prior to solid-phase-extraction was developed to achieve a good recovery of the analytes and to eliminate the interfering species. After derivatization with MSTFA, samples were analyzed by GC/MS. Specificity, accuracy, precision and linearity of the method were evaluated; LOD and LOQ were, respectively, 10ng/25ng for morphine and 5ng/10ng f… Show more

“…A large proportion of these deaths are contributed to drug overdose, which can lead to sudden cardiac and respiratory arrest. Despite that, many of them can be contributed to the mode of drug intake (Gambaro et al 2014;Guerrini et al 2013). The use of opioids is relatively higher in the Southern part of the West Bank (5.7% total in the last 5 years) compared to the Northern (1.5%) and the Center (3%).…”

“…In these samples also the levels of morphine and codeine were evaluated. Moreover we analyzed other seven samples from heroin fatalities belonging to a period of time between 2012 and 2014 (cases [9][10][11][12][13][14][15]. To this end we studied and validated an analytical method suitable to this purpose, using deuterated internal standards in order to enhance sensitivity (Figure 1).…”

“…In a previous study [10], we developed a gas chromatography/mass spectrometry (GC/MS) method to investigate the brain distribution patterns of morphine and codeine, which was then, applied to the determination of their levels in nuclei specimens from 14 fatal cases of suspected heroin overdose. The results of this analysis were subsequently compared to metabolite levels in the blood obtained from the systematic toxicological analysis (STA) [11].…”

“…The techniques that are most widely used to extract fentanyllike compounds from complex/biological matrices are solidphase extraction (SPE) and liquid-liquid extraction (LLE). SPE [18][19][20] and LLE [21][22][23] are reliable and widespread techniques used in the field of chemical-toxicological analysis for detecting of the majority of toxic compounds. Several slightly different protocols for fentanyls extraction were evaluated throughout the literature.…”
