Development and Validation of a Bioanalytical Method for 3′- and 6′-Sialyllactose in Minipig Liver and Kidney Using Liquid Chromatography-Tandem Mass Spectrometry and Its Application to Analysis of Tissue Distribution

Abstract: Breast milk contains human milk oligosaccharides (HMOs), including sialyllactose (SL). SL is composed of sialic acid and lactose, and is divided into 3′-SL and 6′-SL according to the binding position. SL has immunoprotective effects against bacteria and viruses, and acts as a probiotic in the gastrointestinal tract. In this study, we developed a bioanalytical method for simultaneous analysis of 3′-SL and 6′-SL in liver and kidney tissues of Yucatan minipigs using liquid chromatography–tandem mass spectrometry … Show more

“…The recoveries were calculated by the formulae: recovery (%) = (detected amount − original amount)/spiked amount × 100%. The matrix effect refers to the enhancement or suppression of a chromatography signal by interference or co-eluting compounds in the matrix [42]. It was evaluated using a slope comparison method.…”

“…Generally, it is necessary to derivate lactose sialylate so that the derivative has a certain ultraviolet absorption peak before it can be detected normally. Such methods have complicated conditions and high conditions for sample preparation and are not suitable for detecting sialyllactose [ 17 , 18 , 19 , 20 , 21 ].…”
