Development and Validation of a Real-Time PCR for Detection of Pathogenic Leptospira Species in Clinical Materials

Abstract: Available serological diagnostics do not allow the confirmation of clinically suspected leptospirosis at the early acute phase of illness. Several conventional and real-time PCRs for the early diagnosis of leptospirosis have been described but these have been incompletely evaluated. We developed a SYBR Green-based real-time PCR targeting secY and validated it according to international guidelines. To determine the analytical specificity, DNA from 56 Leptospira strains belonging to pathogenic, non-pathogenic an… Show more

“…In Amsterdam (2014–2015) and Nijmegen-Doetinchem (2015), kidneys from all captured rats were both tested by culture as well as by real-time PCR at the NRL. At the NRL, culturing, real-time PCR analyses and Leptospira species determination were performed by previously described methods [27]. Prevalence rates and confidence intervals were determined using R version 3.2.0 [28].…”

“…Once the rodent taxonomy has been defined, accurate identification of associated zoonotic pathogens and/or parasites is required, which requires the use of improved screening methods where possible (e.g. Ahmed et al 2009;Al-Sabi and Kapel 2011). The risk to human health posed by major rodent-borne pathogens such as hantaviruses or Yersinia pestis (the agent of bubonic or pneumonic plague) illustrates the need to clearly identify rodent reservoirs and reservoir complexes (Haydon et al 2002).…”
