Development and maturation of thymic dendritic cells during human ontogeny

Abstract: Thymic dendritic cells (TDC) are dendritic cells situated mainly in the cortico-medullary zone and in the medullary region of the thymus. However, the phenotype of TDC during ontogeny is poorly documented. The aim of this study has been to investigate the development and maturation of TDC during human ontogeny. Immunohistochemical analyses and immunoelectron-microscopic investigation of 21 human thymus specimens have been performed to detect the subtypes of TDC by using various DC-related and DC-development-re… Show more

“…This is the first time that S100 immunoreactivity in the thymic medulla in both isolated and epithelial cells of Hassall's corpuscles has been reported. However, it is possible that a subset of these cells with a more dendritic morphology are similar to the S100-positive dendritic cells described by Uccini et al (1986) and Savchenko et al (2006). Savschenko et al showed that S100-positive dendritic cells are present in the corticomedullary region and the medulla of the human thymus at an early stage of gestation, suggesting that S100-positive cells are candidates for the thymic dendritic cell precursor.…”

“…During early ontogeny (4 months), S100 + thymic dendritic cell precursors populate the corticomedullary border and medullary region of thymus. Other thymic dendritic cell markers such as CD1a and Langerin appear later (at 5 months gestation) [63]. In thymus of newborns S100-protein + cells, including interdigitating dendritic cells, were found in thymic medulla, at the corticomedullary junction, nearby and inside Hassall's corpuscles, but rarely in the cortex [64], [65].…”

“…Immunohistochemical staining was performed both on paraffin and on frozen sections as previously described 21, using various monoclonal antibodies (see the above section entitled ‘Monoclonal antibodies’). Antigen retrieval was performed only for paraffin sections.…”
