Tibouchina mutabilis (Vell.) Cogn. (Melastomataceae) is a characteristic species of Serra do Mar, with potential for recovery of degraded areas and with landscaping purpose. The aim of this study was to investigate the acquisition of germination capacity of seeds and establishment of the best period for fruit harvest by weekly harvests, when morphological and germination capacity features were observed in four stages of development. The T. mutabilis seeds germinated only under white light. Besides the necessity of light for germination, seeds of T. mutabilis did not show another type of dormancy at the end of the ripening, with high germination percentage. The germination rate increases progressively in the function of lose of moisture in the subsequent stages of development, suggesting that the high moisture was the factor that restricted the germination. The best indicator of physiological maturation of seeds is the natural opening of the capsule. The advantage of this parameter is that it can be widely utilized and it is easy to recognize it. Due to the anemochoric dispersion of seeds, we propose that the harvest must be done, with good results, of fruit with 21 days after anthesis, and drying the fruit for capsule opening for seed extraction.Keywords: seed germination, seed maturation, morphology. (Melastomataceae), espécie característica da Serra do Mar, apresenta potencial para recuperação de áreas degradadas e para fins paisagísticos. Objetivou-se investigar a aquisição da capacidade germinativa das sementes e o estabelecimento da melhor época para colheita de frutos da espécie por meio de coletas semanais, onde as características morfológicas e a capacidade germinativa foram observadas, em quatro estádios de desenvolvimento. A capacidade germinativa foi observada com 14 dias pós-antese e a máxima germinação foi observada aos 21 dias. As sementes T. mutabilis germinaram somente na luz. Além da necessidade da luz para a germinação, as mesmas não apresentaram nenhum tipo de dormência na fase final de maturação, apresentando alta porcentagem de germinação, A velocidade de germinação aumentou progressivamente em função do dessecamento das sementes nas subseqüentes fases avaliadas, sugerindo que a alta umidade foi o fator limitante para a germinação. Sesame is an oilseed crop which can be grown on marginal lands. Selection of stable sesame cultivars that can adapt to local environmental conditions can be a very important food security strategy. A set of 21 high yielding sesame accessions that have been selected for a temperate region were grown in the transitional zone of Ghana during the major season of 2014. The seeds were sown after the first rain in the year in a completely randomized design, with three replications. The objective was to evaluate the effect of the contrasting environment on sesame phenology and to select cultivars with yield potential that can be accepted into local farming systems in the new environment. Morphological, physiological and agronomic traits, leading to yield were recorded in this study. Number of capsules per plant had the strongest association (72%) with seed yield. Five accessions showed a combination of early maturity < 12 weeks with high overall mean seed yield (> 20 g per plant) and good harvest index (0.29). Based on their mean performance these cultivars have been selected as promising exotic cultivars for the new locality.

Sesame (Sesamum indicum L. syn. Sesamum orientale L.) is considered to be the first oil seed crop known to man. Despite its versatile use as an oil seed and a leafy vegetable, sesame is a neglected crop and has not been a subject of molecular genetic research until the last decade. There is thus limited knowledge regarding genome-specific molecular markers that are indispensible for germplasm enhancement, gene identification, and marker-assisted breeding in sesame. In this study, we employed a genotyping by sequencing (GBS) approach to a sesame recombinant inbred line (RIL) population for high-throughput single nucleotide polymorphism (SNP) identification and genotyping. A total of 15,521 SNPs were identified with 14,786 SNPs (95.26 %) located along sesame genome assembly pseudomolecules. By incorporating sesame-specific simple sequence repeat (SSR) markers developed in our previous work, 230.73 megabases (99 %) of sequence from the genome assembly were saturated with markers. This large number of markers will be available for sesame geneticists as a resource for candidate polymorphisms located along the physical chromosomes of sesame. Defining SNP loci in genome assembly sequences provides the flexibility to utilize any genotyping strategy to survey any sesame population. SNPs selected through a high stringency filtering protocol (770 SNPs) for improved map accuracy were used in conjunction with SSR markers (50 SSRs) in linkage analysis, resulting in 13 linkage groups that encompass a total genetic distance of 914 cM with 432 markers (420 SNPs, 12 SSRs). The genetic linkage map constitutes the basis for future work that will involve quantitative trait locus (QTL) analyses of metabolic and agronomic traits in the segregating RIL population.
