The development of competitive enzyme immunoassays for ovine plasma LH (oLH) and FSH (oFSH) is described. Standards and plasma samples were preincubated with diluted antiserum to oLH or oFSH and the reacted solution (100 micro l per well) was transferred to plates previously coated with oLH or oFSH, respectively. The second antibody used was anti-rabbit IgG horseradish peroxidase. The measuring range was 0.39-50 ng/ml for each hormone and the 50% relative binding sensitivity was 9 ng/ml for oLH. The respective value for oFSH was 3.5 or 34 ng/ml with different hormone and antibody preparations used for the assay. The enzyme immunoassays were used to determine oLH and oFSH levels in plasma from ewes of two breeds during the oestrous cycle. The assays detected the first FSH surge coincident with the LH surge, the second FSH surge about 24 h later and the periodic fluctuations of FSH concentrations during the luteal phase of the oestrous cycle. These enzyme immunoassays are an efficient and economic alternative to the established radioimmunoassays (RIA) for oLH and oFSH.

Bacterial toxins are widespread in the environment as well as in the digestive system of humans and animals. Toxin from Gram-negative bacteria (endotoxin or lipopolysaccharide; LPS) has a life-long programming effect on reproduction in rats, but the mediators have not been well-documented, so we investigated the effects of LPS on the timing of puberty in female rats. Because the levels of nitric oxide (NO) and interleukin 1β (IL-1β) increase following injection of LPS, we injected neonates (post-natal day (pnd) 7) with LPS, with or without NO or IL-1β inhibitors. Half of the prepubescent (pnd 30) animals received an additional LPS injection. Vaginal opening, number of ovarian follicles, and serum anti-LPS antibodies were determined. A single LPS injection was sufficient to reduce the primordial follicle pool, but puberty was delayed when rats received 2 LPS injections (at pnd 7 and 30). NO or IL-1β inhibitors improved both of these parameters, suggesting that the early detrimental effects of LPS on puberty and primordial follicle pool are mediated by NO and IL-1β.

Background/Aims: The suprachiasmatic nucleus (SCN) is thought to control the timing of luteinizing hormone (LH) surges. The present study was designed to examine temporal patterns of Fos expression in the dorsomedial and ventrolateral parts of the SCN (SCNdm and SCNvl) of female rats during an LH surge. It also included examination of temporal changes in plasma LH levels and temporal changes in Fos levels in the anteroventral periventricular nucleus (AVPV) and gonadotropin-releasing hormone (GnRH) neurons. Methods: Ovariectomized rats injected with 20 µg estradiol benzoate (EB) or vehicle were sacrificed at various times from Zeitgeber time (ZT) 8:00 to 16:30 h (ZT8–16.5; ZT0 = lights on; ZT12 = lights off) on the 2nd day after the injection. Immunohistochemical analyses for Fos and GnRH and enzyme-linked immunosorbent assays for LH were then performed. Results: In both the SCNdm and SCNvl of EB rats, the number of Fos-immunoreactive cells significantly increased between ZT9.5–10.5 and ZT11–12. On the other hand, in EB rats there were significant peaks of LH levels and Fos levels in GnRH neurons and the AVPV between ZT11–12 and ZT13–14. There was no significant difference in the number of Fos-immunoreactive cells between EB and control rats in either the SCNdm or SCNvl at ZT9.5–10.5, or in the SCNdm at ZT11–12, whereas the SCNvl of EB rats contained more Fos-immunoreactive cells than that of control rats at ZT11–12. Conclusion: These results suggest that in female rats during an LH surge, a peak in the Fos level in the SCN precedes peaks in Fos levels in the AVPV and GnRH neurons.
