Micro- and nanoemulsions are potential drug solubilizers and penetration enhancers through the high surfactant/co-surfactant content. This study aimed to evaluate the influence of minoxidil (MXD) solubilized in the microemulsions (MEs) on drug release by in vitro/ex vivo diffusion through the semi-permeable membrane Spectra/Por® (Spectrum Laboratory, Gardena, CA, USA) and porcine ear skin. Moreover, a residual amount of drug in the skin after ex vivo diffusion was evaluated. The reference MER, lecithin-containing MEL, and gelatin-containing MEG were characterized in terms of their size, polydispersity index, density, viscosity, electrical conductivity and surface tension. Based on the in vitro diffusion, it can be argued that MEL slowed down the drug release, while MER and MEG have no significant effect compared to the sample, in which propylene glycol (PG) was used as a solubilizer. Determination of the residual drug amount in the skin after 6 h of the ex vivo permeation was demonstrated as the most valuable method to evaluate the effectiveness of the ME’s application. The results indicate that the most optimal MXD permeation enhancers in alginate gel were the natural surfactants containing MEs. MXD solubilization in MEG and MEL had caused more than 5% of the drug remaining in the skin, which is almost a 1.5-fold higher amount compared to the reference gel.

“Flawless skin is the most universally desired human feature” is an iconic statement by Desmond Morris. Skin indicates one´s health and is so important that it affects a person’s emotional and psychological behavior, these facts having propelled the development of the cosmetics industry. It is estimated that in 2023, this industry will achieve more than 800 billion dollars. This boost is due to the development of new cosmetic formulations based on nanotechnology. Nanocarriers have been able to solve problems related to active ingredients regarding their solubility, poor stability, and release. Even though nanocarriers have evident benefits, they also present some problems related to the high cost, low shelf life, and toxicity. Regulation and legislation are two controversial topics regarding the use of nanotechnology in the field of cosmetics. In this area, the U.S. FDA has taken the lead and recommended several biosafety studies and post-market safety evaluations. The lack of a global definition that identifies nanomaterials as a cosmetic ingredient is a hindrance to the development of global legislation. In the EU, the legislation regarding the biosafety of nanomaterials in cosmetics is stricter. “The cost is not the only important issue, safety and the application of alternative testing methods for toxicity are of crucial importance as well”.

Alopecia can be characterized as a hair-related disorder leading to decreased hair growth, especially on the scalp. It has affected approximately 2% of the world population. The hair loss may emerge due to genetic or environmental factors like stress, infectious agents, tensile forces or medications. The pathogenetic pathway of induction of alopecia comprises of different factors, but exploitation of hair follicle immune privilege system (HFIP) is the most prominent one. Alopecia can be classified into two types: scarring alopecia or non-scarring alopecia. Although, many conventional dosage forms have been formulated against alopecia, but they have been associated with different adverse effects such as weight gain, palpitations, itching etc. In order to overcome the adverse effects associated with conventional dosage forms, the topical nano-formulations have been used for the treatment of dermatological disorders. Nano-formulations are known to increase the contact time of drug with the target site, thereby, achieving sustained and controlled release of the drug leading to enhanced therapeutic compliance. This review provides an in-depth study of developed nanocarriers like liposomes, nano-emulsions, ethosomes, solid lipid nanoparticles (SLNs), nanostructured lipid carriers (NLCs) along with their composition, method of preparation, results obtained after formulation, suitable use and along with their potential use in treatment of different forms of alopecia.
