Determination of Water-Soluble Vitamins B1, B2, B3, B6, B9, B12 and C on ODS Column 5 μm by High Performance Liquid Chromatography with UV-DAD Detection

Abstract: HPLC method was developed and applied to the determination of some water-soluble vitamins (B1, B2, B3, B6, B9, B12 and C) in Syrian pharmaceutical preparations. Reserved phase, ion-pair high performance liquid chromatography with UV-DAD detection was employed. The developed chromatographic method employed an ODS column with dimension (150 × 4.6 mm; 5 µm). The mobile phase was 5.3 mM of hexane-1-sulfonic acid sodium with 0.1 % triethylamine as solvent (A) at pH 3.3 and 5.3 mM of hexane-1-sulfonic acid sodium:ac… Show more

“…Vitamin C (Ascorbic acid), a water-soluble vitamin, was measured according to Antakli et al . 31 , by reversed phase-liquid chromatography with UV detection (λ em 246 nm) using BDS-HYPERSIL-C18 column (100 × 4.6 mm, 3 µm, Thermo Fisher Scientific Inc., USA) with mobile phase of (A): 5.84 mM of hexane-1-sulfonic acid sodium: acetonitrile (95:5, v/v) with 0.1% triethylamine, (B): 5.84 mM of hexane-1-sulfonic acid sodium: acetonitrile (50:50) with 0.1% triethylamine, both 2.5 pH adjusted using 1 M orthophosphoric acid. On the other hand, Vitamin E (α-tocopherol), fat-soluble vitamin, was measured according to Barba et al .…”

“…Vitamin B complexes were fractionated according to the method defined by Antakli et al [ 65 ], with a slight modification using a variable wavelength detector (VWD) instead of a fluorescence detector with the VWD set at 280 nm. This method was validated by following some details, including LOD, LOQ and recovery range (97–100%) [ 65 ].…”

“…A gradient flow was started with 100% of solvent A until the mobile composition, then changed to 50% of A and 50% of B for 5 min. The UV-PDA (photodiode array) wavelength absorbance detector was set at 455 nm (vitamin A), 246 nm (vitamins C and B1), 267 nm (vitamin B2), 260 nm (vitamin B3), 204 nm (vitamin B5), 290 nm (vitamin B6), 282 nm (vitamin B9), 280 nm (vitamin D), and 290 nm (vitamin E) [20,21]. System suitability was considered with respect to instrument precision, resolution, tailing factor, and retention time.…”
