Determination of volatiles, antioxidant activity, and polyphenol content in the postharvest waste of Ocimum basilicum L

Hydrodistillation (HD) systems have been popular for essential oils extraction from medicinal and aromatic plants for several decades. Nevertheless, they substantially consume thermal or electrical energy and have a visible carbon footprint on the environment. This article describes a state‐of‐the‐art review of the HD systems used for essential oil extraction to examine their effectiveness. From this study, it has been found that, for a microwave‐assisted system, the yield of an essential oil varies from 0.079% to 8.62%. The microwave‐assisted hydrodistillation (MAHD) system yields 32% more than a conventional HD system. The average extraction time saved using the MAHD system is 67.17% compared with the traditional one. Ohmic‐assisted HD system is better than the microwave‐assisted and conventional HD system, with an 18.04% enhancement in the yield. During this study, it is also investigated that for the distillation system operating at atmospheric pressure, the extraction temperature ranges from 80°C to 120°C. This temperature can easily be attained in a solar hydrodistillation system. Several investigators have demonstrated the use of solar energy for essential oil extraction. However, these systems mostly use a concentrating collector, henceforth found bulky and expensive.

An optimization procedure for multistep temperature-programmed capillary gas chromatography was developed for the analysis of basil essential oil. The current study was planned after performing the following three steps: a preliminary chromatographic study, primary parameter screening, and final method optimization by using 23 factorial and Doehlert designs (DOE). The optimized conditions were obtained by using a split ratio of 1 / 2 , a gas flow rate of 1.3 mL/min, and a heating rate of 2°C/min. This set of conditions was later used for the optimization of the final method. The optimal experimental conditions were subsequently obtained by response surface optimization (isotherm duration = 6.2 min and slope = 1.06°C/min). The compositions of essential oils from six varieties of basil (Ocimum B. (1), Ocimum B. purpurescens opal (2), Ocimum B. minimum (3), Ocimum. B. cinnamon (4), Ocimum B. (marcellas) (5), and Ocimum B. Citriodora (6)) grown in four regions of Algeria (Ouargla, south of Algeria (O); Mostaganem, west of Algeria (M); Algiers, north of Algeria (A), and Constantine, east of Algeria (C)) were determined. A statistical study was performed based on hierarchical ascending classification and principal component analysis to elucidate the relationships between the basil varieties, their region of growth, and their essential oil composition. In addition to identifying several chemotypes, such as linalool, linalool/eugenol, linalool acetate, methyl eugenol, methyl chavicol, eugenol, methyl cinnamate E, and geraniol, a new compound, namely, octadecenamide (Z) (oleamide), was detected in the essential oil of Ocimum basilicum L. from Algiers (A1).

This study aimed to determine the chemical composition of the essential oils (EOs) of Ocimum basilicum L., as well as to evaluate the antibacterial, antidiabetic, dermatoprotective, and anti-inflammatory properties, and the EOs and aqueous extracts of O. basilicum. The antibacterial activity was evaluated against bacterial strains, Gram-positive and Gram-negative, using the well diffusion and microdilution methods, whereas the antidiabetic activity was assessed in vitro using two enzymes involved in carbohydrate digestion, α-amylase and α-glucosidase. On the other hand, the dermatoprotective and anti-inflammatory activities were studied by testing tyrosinase and lipoxygenase inhibition activity, respectively. The results showed that the chemical composition of O. basilicum EO (OBEO) is dominated by methyl chavicol (86%) and trans-anethol (8%). OBEO exhibited significant antibacterial effects against Gram-negative and Gram-positive strains, demonstrated by considerable diameters of the inhibition zones and lower MIC and MBC values. In addition, OBEO exhibited significant inhibition of α-amylase (IC50 = 50.51 ± 0.32 μg/mL) and α-glucosidase (IC50 = 39.84 ± 1.2 μg/mL). Concerning the anti-inflammatory activity, OBEO significantly inhibited lipoxygenase activity (IC50 = 18.28 ± 0.03 μg/mL) compared to the aqueous extract (IC50 = 24.8 ± 0.01 μg/mL). Moreover, tyrosinase was considerably inhibited by OBEO (IC50 = 68.58 ± 0.03 μg/mL) compared to the aqueous extract (IC50 = 118.37 ± 0.05 μg/mL). The toxicological investigations revealed the safety of O. basilicum in acute and chronic toxicity. The finding of in silico analysis showed that methyl chavicol and trans-anethole (main compounds of OBEO) validate the pharmacokinetics of these compounds and decipher some antibacterial targets.
