Determination of uric acid on continuous-flow (AutoAnalyzer II and SMA) systems with a uricase/phenol/4-aminophenazone color test.

Abstract: This report describes a new specific colorimetric procedure for uric acid assay with AutoAnalyzer II and SMA (Technicon) systems, made specific by the application of uricase. Hydrogen preroxide, formed in this reaction, effects the oxidative coupling of 4-aminophenazone and 2,4-dichlorophenol under the catalytic influence of peroxidase. The red dye formed is measured at 505 or 520 nm. A sample blank measurement is not necessary, and the reagents show very good stability. The test shows linearity up to 714 mumo… Show more

“…The urate oxidase activity was determined using a modified urate oxidase/phenol/4-aminoantipyrine colorimetric assay (Klose et al 1987 )). Briefly, uric acid can be specifically converted to allantoin and hydrogen peroxide by urate oxidase.…”

“…Uricase activity was assayed by a HRP-coupled reaction using horseradish peroxidase (10 mM), uric acid (1.5 mM), 4-AAP (12 mM), and phenol (60 mM) in 20 mM potassium phosphate buffer (pH 7.0) (Klose et al 1978). The amount of produced H 2 O 2 was measured using HRP catalyzing the oxidation of phenol in the presence of 4-AAP (Asad et al 2014).…”
