Determination of triamcinolone in human plasma by a sensitive HPLC–ESI‐MS/MS method: application for a pharmacokinetic study using nasal spray formulation

Abstract: A liquid chromatography-electrospray ionization tandem mass spectrometry (HPLC-ESI-MS/MS) method for the quantitation of triamcinolone in human plasma after nasal spray application was developed and validated. Betamethasone was used as internal standard (IS). The analytes were extracted by a liquid-liquid procedure and separated on a Zorbax Eclipse XDB C(18) column with a mobile phase composed of 2 mM aqueous ammonium acetate pH 3.2 and acetonitrile (55:45). Selected reaction monitoring was performed using the… Show more

“…Immediately after withdrawal, 1-μl samples were diluted with 50 μl mobile phase consisting of acetonitrile/2 m m aqueous ammonium acetate (60:40) adjusted to pH 3.2 with formic acid [ Cesar et al, 2011 ] and stored at +4°C until analysis by high-performance liquid chromatography/mass spectrometry, comprising an Ultimate RSLC 3000 series System (Thermo Fisher Scientific, Vienna, Austria) and an API 4000 Triple Quadrupole Mass Spectrometer (AB Sciex instruments, Vienna, Austria) equipped with an electrospray ionization ion source and controlled by the Analyst 1.5 software (Dionex, Vienna, Austria). The chromatographic separation of 5-µl samples was conducted by isocratic elution using an Acclaim® 120 C 18 reversed-phase LC column (2.1 × 150 mm, 3 µm; Thermo Fisher Scientific, Vienna, Austria) at 25°C.…”

“…The steroid profile included 21-DF, D4A, testosterone, and 17-OHP. The potential presence of exogenous glucosteroids in DBS was detected using the multiple reaction monitoring (MRM) fragmentation transitions m/z 435 → 415 and m/z 393 → 373 for triamcinolone acetonide and betamethasone, respectively ( 16 ).…”

“…The TA concentrations in rat plasma or joint tissue were determined using the LC–MS/MS assay previously reported [23]. In brief, thawed plasma (100 μL) was mixed with 900 μL of methanol and vigorously vortexed for 10 min, to precipitate protein.…”
