Determination of tissue plasminogen activator and its "fast" inhibitor in plasma.

Abstract: We describe efficient, accurate methods for specific determination of tissue plasminogen activator (t-PA, EC 3.4.21.31) and its "fast" inhibitor in plasma. In this coupled assay, a sample containing t-PA is incubated with plasminogen, a plasmin (EC 3.4.21.7) substrate of low Km and high Kcat, and fibrin as a stimulator. The inhibitor of t-PA is determined by incubating the sample with a known amount of t-PA in excess, then determining the residual t-PA. Both t-PA and t-PA inhibitor can be determined in many sa… Show more

“…Fibrinogen was measured according to the Clauss (1957) method (Fibri-Prest Aut from STAGO, Asnieres-sur-Seine, France) with a total coefficient of variation (CV) of 4% at 1·7 g / l and 5% at 2·6 g / l. An enzyme-linked immunosorbent assay (ELISA; Coalisa-tPA, Haemochrom Diagnostica, Mölndal, Sweden; Holvoet et al, 1985) with a total CV of 14% was used to measure t-PA antigen. PAI-1 activity was determined using Spectrolyse (Biopool, Umeå, Sweden; Chmielewska & Wiman, 1986) with a total CV of 13%. IGF-I was determined by a radioimmunoassay as described previously (Leonsson et al, 2002).…”

“…The ELISA (MacGregor & Booth, 1988) was modified as described previously . PAI activity assay was based upon neutralization of t-PA (Chmielewska & Wiman, 1986). T-PA antigen was assayed by ELISA (MacGregor et al, 1987).…”

“…PAI-1 antigen and activity and t-PA antigen were determined on citrated plasma samples (Chmielewska & Wiman, 1986). For determinations of t-PA activity plasma from blood sampled in commercially available tubes containing acidified citrate (Biopool AB, Umeta, Sweden) was used (Chmielewska & Wiman, 1986). For…”
