Determination of the glycosylation site of flavonoid monoglucosides by metal complexation and tandem mass spectrometry

Abstract: Metal complexation and tandem mass spectrometry were used to differentiate C-and O-bonded flavonoid monoglucoside isomers. Electrospray ionization of solutions containing a flavonoid glycoside and a metal salt led to the generation of the key [M(II) (L) (L-H)] ϩ complexes, where M is the metal ion and L is the flavonoid glycoside. Thirteen flavonoid monoglucosides were examined in combination with Ca(II), Mg(II), Co(II), Ni(II), and Cu(II). Collisional activated dissociation (CAD) of the [M(II) (L) (L-H)] ϩ co… Show more

“…Comparing the values obtained for the solvent molecules with those reported in Table 6 (dE 2 ) for the most stable (4). These results suggest a reasonable explanation for the different charged species observed in the mass spectra of both isomers, i.e., doubly charged species are the most important among the calcium complexes of 2 while they are not observed for 1 (Figure 2a).…”

“…Finally, relative stability of doubly charged species upon deprotonation by a neutral molecule generating two singly charged molecules was analyzed, i.e., (2), (3), (4). Possible relevant neutral molecules are the neutral complexes as well as the solvent molecules present in the source, namely methanol and water.…”

“…We have developed several methods for identifying the conjugation positions and saccharide identities of flavonoid glycosides using tandem mass spectrometry [23][24][25][26][27][28][29]. These methods exploit the natural chelating ability of flavonoids to form metal complexes in solution, which can then be analyzed in a mass spectrometer for structural identification.…”

“…Although flavonoids deprotonate efficiently, the resulting fragmentation patterns often do not give diagnostic ions adequate for differentiation of isomers. Metal complexation is an alternative ionization strategy that has been explored in our group for improved analysis of flavonoids [17][18][19][20][21][22][23], and we have explored metal complexation both with and without the use of an auxiliary …”
