Determination of the complex of cell wall substances in plant products

Abstract: The authors present a new method for the determination of the complex of vegetable cell wall substances. The sample is extracted with boiling 80 % ethanol, boiling absolute ethanol and cold water. The residue corrected for ash, protein, and, if necessary, for starch, gives the amount of cell wall substances. Determinations were made of the same samples of which Salo in this department, using quite a different principle, has determined the cell wall complex. She determined separately cellulose, neutral sugar he… Show more

“…The primary problems associated with NFE and CF fractions (Van Soest 1994) were realised by Paloheimo (1953), who initiated research to develop improved analytical methods for plant cell wall. In the pioneering work, Paloheimo and co-workers (Paloheimo andPaloheimo 1949, Paloheimo andVainio 1965) used a weak hydrochloric acid and a two-stage ethanol extraction to remove cellular contents to describe vegetable fibre. Despite the correct criticism against fractionating feed carbohydrates into CF and NFE, these methods were too laborious, not applicable to faecal samples and the fibre residue was contaminated with protein.…”

“…From the meals and ground rusks determinations were made of dry matter, ash and crude protein. Plant cell wall substances were also determined by the methods of Salo (1965 a) and Paloheimo and Vainio (1965), and Cr 2 0 3 from the rusks (Paloheimo and Paloheimo 1935).…”

“…In the studies carried out by Rubner (1916aRubner ( , 1916Rubner ( b, 1919 at the beginning of this century, on the other hand, results approximating rather closely the total amounts of cell-wall material were achieved, the methods are very complex for routine work, however. The procedure of Paloheimo and Vainio (1965), requiring differentcorrection determinations, is also laborious, even though it gives a rather accurate determination of the complex of cell-wall substances in animal foods.…”
