a b s t r a c tNon-alcoholic fatty liver disease (NAFLD) is an increasingly reported pathology, characterized by fat accumulation within the hepatocyte. Growing evidences suggest specific effects on mitochondrial metabolism, but it is still unclear the relationship between fatty liver progression and mitochondrial function. In the present work we have investigated the impact of fatty liver on mitochondrial bioenergetic functions and susceptibility to mitochondrial permeability transition (MPT) induction in animals fed a choline-deficient diet (CDD) for 4, 8, 12 or 16 weeks. Mitochondria isolated from CDD animals always exhibited higher state 4 respiration. Mitochondrial membrane potential was decreased in CDD animals at 4 and 16 weeks. At 12 weeks, oxidative phosphorylation was more efficient in CDD animals, suggesting a possible early response trying to revert the deleterious effect of increased triglyceride storage in the liver. However, mitochondrial dysfunction was evident in CDD animals at 16 weeks as indicated by decreased RCR and ADP/O, with a corresponding decrease in respiratory chain enzymes activities. Such loss of respiratory efficiency was associated with accumulation of protein oxidation products, in tissue and mitochondrial fraction. Additionally, although no differences in ATPase activity, the lag phase was increased in mitochondria from CDD animals at 16 weeks, associated with decreased content of the adenine nucleotide translocator. Increased susceptibility to calcium-induced MPT was evident in CDD animals at all time points. These results suggest a dynamic mechanism for the development of NALFD associated with altered mitochondrial function.

RNA PROCESSING FACTOR1 (RPF1) and RPF2 are pentatricopeptide repeat (PPR) proteins involved in 5# processing of different mitochondrial mRNAs in Arabidopsis (Arabidopsis thaliana). Both factors are highly similar to RESTORERS OF FERTILITY (RF), which are part of cytoplasmic male sterility/restoration systems in various plant species. These findings suggest a predominant role of RF-like PPR proteins in posttranscriptional 5# processing. To further explore the functions of this group of proteins, we examined a number of T-DNA lines carrying insertions in the corresponding PPR genes. This screening identified a nearly complete absence of mature ccmC transcripts in an At1g62930 T-DNA insertion line, a phenotype that could be restored by the introduction of the intact At1g62930 gene into the mutant. The insertion in this nuclear gene, which we now call RPF3, also leads to a severe reduction of the CcmC protein in mitochondria. The analysis of C24/rpf3-1 F2 hybrids lacking functional RPF3 genes revealed that this gene has less influence on the generation of the mature ccmC 5# transcript end derived from a distinct ccmC 5# upstream configuration found in mitochondrial DNAs from C24 and other accessions. These data show that a particular function of an RF-like protein is required only in connection with a distinct mtDNA configuration. Our new results further substantiate the fundamental role of RF-like PPR proteins in the posttranscriptional generation of plant mitochondrial 5# transcript termini.

RESUMENAntecedentes: El linalool (2,6-dimetil-2,7-octadien-6-ol) es un monoterpeno presente en más de 200 tipos de plantas, y constituye el componente mayoritario de varios aceites esenciales. El linalool posee una amplia variedad de potenciales propiedades farmacológicas; que van desde propiedades antisépticas, ansiolíticas, anti-inflamatorias, antifúngicas, antimicrobianas y proapotóticas en diferentes líneas de cé-lulas tumorales. Sin embargo, su principal uso está relacionado como una molécula odorante o fragancia en productos de uso tópico en productos cosméticos y en la industria del perfume. Objetivo: En este estudio se evaluó el efecto del linalool sobre la bioenergética de mitocondrias aisladas de hígado de rata. Métodos: El linalool en concentraciones entre (5-100μg/mL) fue evaluado en mitocondrias aisladas de ratas machos albinas raza Wistar, de acuerdo al método descrito por Voss et al (1961). Inicialmente, se determinó el consumo de oxígeno en mitocondrias intactas mediante un método polarográfico por medio de un oxígrafo. Posteriormente, se determinó el efecto del compuesto sobre las enzimas de la cadena respiratoria mitocondrial usando métodos polarográficos y espectrofotométricos. Resultados: Los resultados obtenidos indican que el linalool en las concentraciones de 5, 10, 50 y 100μg/mL, afecta la velocidad de consumo de oxígeno en el estado 3 y 4 de mitocondrias energizadas con glutamato-malato y succinato. Sin embargo, el efecto inhibitorio es mayor al usar glutamato-malato como sustrato. Por su parte, el análisis de la actividad de las enzimas de la cadena respiratoria mitocondrial mostró que el linalool (50μg/mL) inhibe los complejos I, III y la actividad ATPasa. Conclusiones: El linalool inhibe el complejo I, complejo III y la ATPasa, esta inhibición podría aumentar la generación de especies reactivas de oxígeno.Palabras clave: Linalool, mitocondria, enzimas, consumo de oxígeno.
