Determination of serotonin, melatonin and metabolites in gastrointestinal tissue using high‐performance liquid chromatography with electrochemical detection

Abstract: In this paper we show a simple isocratic chromatographic method for the detection of serotonin and its precursors and metabolites from various types of gastrointestinal tissue. The paper measures for the first time basal measurements of melatonin in the gastrointestinal tract, which has recently been shown to be released from the musosal lining of the gut. Tissue samples were stable following sample preparation in either 0.1 m perchloric acid or mobile phase. Analysis was carried out using a mobile phase consi… Show more

“…This detection limit is significantly lower than that reported in other works for 5-HT using differential pulse voltammetry with bare carbon fiber (1.05 μM) and boron doped diamond (3.08 μM) microelectrodes 44. This is also lower than the detection limit obtained using high-liquid performance chromatography with electrochemical detection (241 nM) 29. The low detection limit and high sensitivity of our sensors allowed us to use this technology for determining 5-HT levels in the digestive system of intact embryos, where the concentrations are low.…”

“…For the evaluation of electroactive surface area of the working electrode among different common benchmark redox systems, K 4 [Fe(CN) 6 ] was selected for its surface sensitive electrochemical response, especially for carbonaceous materials. 3-redox pair present in solution in anodic and cathodic scans.…”

“…3,4 Numerous analytical methods were reported for detection and quantification of melatonin, such as ultraviolet-visible (UV-vis) spectrophotometry, chemilumenescence, fluorometry, thin-layer chromatography, gas chromatography-mass spectrometry, and high-performance liquid chromatography. [5][6][7][8][9] Furthermore, melatonin is an antioxidant compound that can be analyzed with electrochemical methods. 10 Voltammetric and amperometric methods were applied for the determination of melatonin using carbon paste electrodes, carbon disk electrode, carbon fiber microdisk electrode, etc.…”

“…This is expected as shown in Scheme 1 only 5-HT is released from the EC cell and thus can be monitored. At present the majority of assays studying the efficacy of the transmission process rely on monitoring homogenised tissue samples, which can only monitor intracellular components [18,28,30]. Therefore assumptions about the transmission mechanism are being made based on what can be monitored and thus for example intracellular 5-HT is often used as a marker of release, even if only a fraction of this is actually released [33,34].…”

“…However, in contrast, high performance liquid chromatography (HPLC) [18,29,30] is capable of measuring all parameters noted in Scheme 1. Within the field of gastroenterology, HPLC has been predominantly used to measure levels of analytes in tissue homogenates [18,28,30] and hence only intracellular neurochemicals are monitored, meaning the release of 5-HT from EC cells is entirely missed. On the other hand, there are methods such as microdialysis [31,32] that only measure extracellular levels of 5-HT and so intracellular neurochemicals are not taken into account.…”
