Determination of Plant Thiols by Liquid Chromatography Coupled with Coulometric and Amperometric Detection in Lettuce Treated by Lead(II) Ions

Abstract: The main aim of this paper is to utilize high performance liquid chromatography with electrochemical detection for determination of thiols content in plants tissues of lettuce treated with lead(II) ions (0, 0.5 and 1 mM). We used two HPLC-ED instruments: HPLC coupled with one channel amperometric detector and HPLC coupled with twelve channel coulometric detector to detect simultaneously twelve thiols. The detection limits of thiols measured by CoulArray detector were about two magnitudes lower in comparison to… Show more

“…More- Because reduced glutathione is a substrate for PCS and based on the abovementioned optimized conditions, the following chromatographic parameters were selected. As previously published [22,24,25], these parameters were used for detection of GSH, GSSG and PC 2 simultaneously with the highest sensitivity for PC 2 . Both the detector and the column were thermostatically controlled at 30 • C. Mobile phase consisted of A: trifluoroacetic acid (80 mM) and B: 100% methanol.…”

“…In contrast, electrochemical detection (ED) is an attractive alternative method for electroactive species detection, because of its inherent advantages of simplicity, ease of miniaturization, high sensitivity and relatively low cost. If electrochemical detector is coupled with effective separation method such as high performance liquid chromatography (HPLC) [22][23][24][25][26][27][28][29][30], capillary electrophoresis [31,32] and/or capillary liquid chromatography [33], real sample can analysed with high sensitivity and selectivity.…”

“…For instance, metallothioneins, DL-homocysteine, N-acetyl-L-cysteine, γ-glutamylcysteine, homoglutathione and PC have also been found in soil plant tissues (Alvarez-Legorreta et al, 2008;Cobbett and Goldsbrough, 2002;Diopan et al, 2010;Klapheck, 1988). P. australis leaves presented in general significantly higher amounts of NPT and total thiols than in roots.…”

“…Glutathione is the most abundant and important low molecular mass thiol, widely distributed in living cells and involved in many biological reactions such as maintaining the homeostasis of heavy metal ions inside the plant cell (Ridnour et al 1999;Diopan et al 2010). Tree-specific accumulation of transcripts in the same forest area from PbGSTZ genes, in fresh leaves, was examined by Northern blot analysis (figure 6).…”
