Two spectrophotometric methods are proposed for the assay of oxcarbazepine (OXC) in bulk and dosage forms using Folin-Ciocalteu phenol reagent (FCP) and 3-methyl-2-benzothiazolinone hydrazine hydrochloride (MBTH) as reagents. The first method involves addition of FCP reagent to OXC in alkaline medium followed by measurement of absorbance at 760 nm (method A), and the other involves addition of a fixed volume of MBTH after treatment of OXC with ferric chloride and measurement of absorbance at 456 nm (method B). In both methods, the amount of chromogen formed corresponds to the amount of OXC and the measured absorbance was found to increase linearly with the concentration of OXC, which is corroborated by the correlation coefficients of 0.9985 and 0.9984 for method A and B, respectively. The systems obey Beer's law for 5-30 mg mL -1 and 10-50 mg mL -1 for methods A and B, respectively. The apparent molar absorptivity was calculated to be 8.06´10 3 L mol -1 cm -1 and 3.126´10 3 L mol -1 cm -1 for methods A and B, respectively. The limits of detection (LOD) and limit of quantification (LOQ) were calculated to be 1.6 and 5 mg mL -1 for method A and 3 and 10 mg mL -1 for method B. The inter-day and intra-day imprecision of the methods were found to be in the range of 1.1-1.7 and 0.9-1.1% for method A, and 1.1-1.9 and 0.6-0.9% for method B. The accuracy ranged between 98.9-99.7% and 99.3-100.1 for method A and B, respectively. No interference was observed from common pharmaceutical excipients. The methods were successfully applied to the assay of OXC in tablet preparations.

Simple, fast and reliable spectrophotometric methods were developed for determination of Oxcarbazepine in bulk and pharmaceutical dosage forms. The solutions of standard and the sample were prepared in Distilled Water. The quantitative determination of the drug was carried out using the zero order derivative values measured at 256 nm and the area under the curve method values measured at 252-258 nm (n=2). Calibration graphs constructed at their wavelengths of determination were linear in the concentration range of Oxcarbazepine using 5-25 μg/ml (r²=0.998 and r²=0.9986) for zero order and area under the curve spectrophotometric method. All the proposed methods have been extensively validated as per ICH guidelines. There was no significant difference between the performance of the proposed methods regarding the mean values and standard deviations. Developed spectrophotometric methods in this study are simple, accurate, precise and sensitive to assay of Oxcarbazepine in tablets.

A simple and rapid ultra-high performance liquid chromatographic (UHPLC) method for the separation and determination of oxcarbazepine and its related substances in tablets, was developed. Chromatographic separation of oxcarbazepine from its related substances (degradation and by-products) was achieved on a reversed phase C 18 column (Pinnacle DB C18, RESTEK, 100 x 2.1 mm, 1.9 µm particle size) using gradient program with water (as mobile phase A) and acetonitrile (as mobile phase B), at a flow rate 0.5 mL min-1 and UV detection at 254 nm. The column temperature was 30°C.The method has good selectivity towards oxcarbazepine and its related substances. The accuracy of the method for oxcarbazepine assay, expressed as mean recovery was 101.8 %, and of the method for quantification of degradation products was 95.9 %-103.3 %. Limit of quantification for oxcarbazepine and its degradation products ranged from 0.60 µg mL-1 to 1.30 µg mL-1. The linearity range for oxcarbazepine assay was from 2.4 to 3.60 mg mL-1 (R 2 =0.999). Mean value for oxcarbazepine assay from the tablets was 299.92 mg (label claim 300 mg). The method can be used for routine analysis and the quality control of oxcarbazepine drug substance and its formulated products.
