Determination of Nicotine in Serum by TLC-Densitometry

Abstract: A TLC-Densitometric procedure for determination of nicotine in serum is reported. Nicotine was extracted with ethylacetate from protein free serum almost quantitatively and evaporated to dryness with a stream of nitrogen. The residue was oxidized to nicotinic acid with concentrated nitric acid, diluted with methanol to a standard volume and then determined quantitatively by TLC-densitometer at X m a r 2 6 2 m. 4a 5

“…It constitutes about 93−94.5% of the total alkaloid fraction in tobacco smoke; the other alkaloids are anatabine 2.7−3.9%, nornicotine 0.96−2.4%, and anabasine 0.35−0.5%, depending on the type of tobaccos . We are interested in applying the optode technique to detect tobacco alkaloids (nicotine as the example of a guest molecule) although many reactions and various techniques have been developed for the determination of nicotine. There is still need for the development of a new method for multipurpose determination of nicotine and other alkaloids.…”
