Determination of gymnemic acid level in Gymnema inodorum leaves using multiple reaction monitoring mass spectrometry

Abstract: Gymnemic acid (GA) is known as the antidiabetic phytoconstituent in Gymnema species, and is used for quality control and standardization of Gymnema products. In Thailand, a number of nutraceutical products of Gymnema inodorum (Lour.) Decne. (GI) are commercially available and this number is increasing. However, standardized GA content information and safety usage guidelines for GI have not been published. The aim of this study was to investigate the amount of GA constituent in GI leaves using mass spectrometry… Show more

“…The validation of LC-ESI-MS/MS with SRM mode for quantifying phenolics in the samples followed the ICH Q2B guidelines (1997) [ 38 ] and works of Srinuanchai et al (2019, 2021) [ 39 , 40 ], to observe parameters such as specificity, linear regression equation, correlation coefficient, limit of detection (LOD), limit of quantification (LOQ), precision, and accuracy ( Table S2 ).…”

“…Linear range, linear regression, correlation coefficients, limit of quantitation (LOQ), limit of detection (LOD), and relative standard deviation (RSD) of the standards were analyzed according to the protocol of Srinuanchai et al 2019 [ 57 ] as shown in Table S3 . The LOQ and LOD were analyzed from the linear calibration curve with the equation as follows: y = a + bx , where y is an area under the peak, a is a y -intercept, b is a slope of the calibration curve, and x is a standard concentration.…”

“…Limit of detection (LOD), limit of quantitation (LOQ), and precision of HPLC standards were shown in Supplementary Table S2 . The validation conditions of each parameter were carried based on the protocol of Srinuanchai et al 2019 [ 63 ]. LOD and LOQ were determined from the linear calibration curve, which expressed as the following equation: y = a + bx where y is an area under the peak, x is a standard concentration, a is a y -intercept, and b is a slope of the calibration curve.…”
